Description

The Tie receptor ligands known as angiopoietins compose a family of proteins. Currently, four angiopoietins have been identified: angiopoietins 1, 2, 3, and 4 (Ang1, 2, 3, and 4). The interaction between Ang1 and the Tie-2 receptor triggers the receptor's autophosphorylation, activating downstream signaling pathways. This crucially contributes to the formation, maturation, and stabilization of blood vessels. Ang2, on the other hand, acts as an endothelium-specific growth factor that acts in opposition to Ang1. It promotes proinflammatory processes associated with vasculature, causing destabilization of quiescent endothelium, vascular leakage, and remodeling. In tumor tissues, Ang2 is selectively expressed and, when combined with other growth factors like VEGF, it facilitates vascular remodeling, angiogenesis, and inflammation.

Background

Binds to actin on the surface of endothelial cells; once bound, angiogenin is endocytosed and translocated to the nucleus. Stimulates ribosomal RNA synthesis including that containing the initiation site sequences of 45S rRNA. Cleaves tRNA within anticodon loops to produce tRNA-derived stress-induced fragments (tiRNAs) which inhibit protein synthesis and triggers the assembly of stress granules (SGs). Angiogenin induces vascularization of normal and malignant tissues. Angiogenic activity is regulated by interaction with RNH1 in vivo.
