Overview | |
Description | One of the members of the CAMKL protein kinase family is MARK4, which is typically found in neural progenitors. In cases of gliomas, however, it is often re-expressed. |
Other names | FLJ90097 is also known as KIAA1860 and is a gene encoding for microtubule affinity regulating kinase 4. It is also referred to as MAP / microtubule affinity regulating kinase 4L. Another variant of this gene is called MAP / microtubule affinity regulating kinase like 1. Additionally, it is sometimes called MAP/microtubule affinity regulating kinase 4 or MAP/microtubule affinity regulating kinase like 1. It is also commonly known as MARK 4 or MARK 4L, and MARK L1. The protein encoded by this gene is a serine/threonine protein kinase known as MARK4. Another name for this protein is MARK4 serine / threonine protein kinase or MARK4 serine/threonine protein kinase. It is sometimes referred to as MARK4_HUMAN or MARK4S. Another name for this protein is MARKL 1 or MARKL1. Additionally, it is associated with the putative protein product of Nbla00650. |

Background | This protein kinase, known as a serine/threonine-protein kinase, has been found to be involved in a range of cellular activities including regulation of the microtubule network, cell cycle progression and energy homeostasis. It is responsible for phosphorylating a number of microtubule-associated proteins, including MAPT, MAP2 and MAP4, which results in the reorganization of microtubules into bundles. Additionally, it is required for the initiation of axoneme extension during cilium assembly. This protein kinase also plays a role in regulating ODF2 localization and phosphorylates the protein in vitro. Along with these cellular functions, the protein kinase can have a negative effect on neuronal cell survival and promote adipogenesis by activating JNK1. However, it can also trigger apoptosis by activating the JNK1 pathway. Interestingly, it has been found to act as a negative regulator of the mTORC1 complex by phosphorylating RPTOR and disrupting the interaction between phosphorylated RPTOR and the RRAGA/RRAGC heterodimer, which is required for mTORC1 activation. |
Tissue specificity | Ubiquitous. Isoform 2 is brain-specific (PubMed:11326310). Expressed at highest levels in brain and testis. Also expressed in heart, lung, liver, muscle, kidney and spleen (PubMed:14594945). |
Posttranslational modification | Ubiquitinated with 'Lys-29'- and 'Lys-33'-linked polyubiquitins which appear to impede LKB1-mediated phosphorylation. Deubiquitinated by USP9X.Phosphorylated at Thr-214 by STK11/LKB1 in complex with STE20-related adapter-alpha (STRADA) pseudo kinase and CAB39 (PubMed:14976552). Phosphorylated throughout the cell cycle (PubMed:25123532). |
