Overview

Description

K16 is a type I cytoskeletal keratin that plays a crucial role in maintaining the structural integrity of epithelial cells. These intermediate filament proteins are further classified into cytokeratins and hair keratins. Cytoskeletal and microfibrillar keratin come in two types: type I and type II. Acidic type I keratin, such as K16 (with a molecular weight of 40-55 kDa) requires a basic counterpart to form filaments. K16 is usually associated with K6. However, in pathological scenarios such as metaplasias, carcinomas of the uterine cervix, and psoriasis vulgaris, K16 and K17 are found to be coexpressed. Hence, K16 plays a critical role in maintaining the stability and strength of epithelial cells under normal conditions as well as in pathological situations.

Other names

Keratin 16, also known as CK-16 or CK16, is a protein encoded by the KRT16 gene in humans. It is a cytoskeletal protein belonging to the keratin family. One of its alternative names is keratin type I cytoskeletal 16. K16 is primarily found in epithelial cells of the skin, nails, and hair. It plays a crucial role in maintaining the structural integrity of these tissues.
Mutations in the KRT16 gene are associated with various skin disorders, including focal non-epidermolytic palmoplantar keratoderma (FNEPPK). FNEPPK is characterized by the thickening of the skin on the palms of the hands and soles of the feet, leading to a hardened and callused appearance. This condition is caused by abnormalities in the keratin proteins, particularly K16.
The expression of K16 can also be induced in certain pathological conditions, such as wound healing and some types of cancers. In these cases, K16 may contribute to tissue repair and participate in cell migration and proliferation.
Overall, studying K16 and its role in skin diseases provides valuable insights into the molecular mechanisms underlying various skin disorders. The understanding of K16's functions and its involvement in disease pathology has the potential to facilitate the development of targeted therapies for these conditions.

Specificity

The detection of endogenous levels of total Keratin 16 can be accomplished using the Keratin 16 antibody. This antibody is designed to specifically target and identify Keratin 16 molecules present in the organism. By using this antibody, researchers can accurately measure and evaluate the levels of Keratin 16 protein in various biological samples.

Application

For Western blot analysis, dilute the primary antibody at a range of 1:500 to 1:3000. Meanwhile, for immunohistochemistry, the antibody should be diluted at a ratio of 1:50 to 1:200. In the case of IF/ICC, the recommended dilution ratio is 1:100-1:500. Finally, for ELISA (peptide), dilute the antibody at a ratio of 1:20000-1:40000. Remember to follow these guidelines to ensure optimal results.

Stability & Storage

It is recommended to store the item at -20 °C for a period of one year. It is important to avoid subjecting it to repeated freeze/thaw cycles.

Storage buffer

This is a description of Rabbit IgG, which is suspended in phosphate buffered saline with a pH of 7.4, 150mM NaCl, 0.02% sodium azide, and 50% glycerol. It is recommended to keep it stored at -20 °C and it will remain stable for 12 months from the time of receipt.

Target

Background

This specific type I keratin plays a critical role in maintaining the integrity of the skin as an epidermis-specific protein. It also acts as a regulator of innate immunity in response to any breach in the skin barrier. Without this protein, various inflammatory checkpoints required for skin barrier maintenance cannot function efficiently. Therefore, its presence is crucial for the protection of our skin against various external factors.

Tissue specificity

Expressed in the hair follicle, nail bed and in mucosal stratified squamous epithelia and, suprabasally, in oral epithelium and palmoplantar epidermis. Also found in luminal cells of sweat and mammary gland ducts.
