Description

IL-27 is a cytokine that exhibits both pro-inflammatory and anti-inflammatory properties. It plays a crucial role in regulating the development of T helper cells and has various effects on innate immune cells. The primary target cells of IL-27 are CD4 T helper cells, which can differentiate into different types of effector cells, including TH1, TH2, and TH17.
One of the key functions of IL-27 is its ability to suppress T-cell proliferation and induce cytotoxic T-cell activity. It also plays a role in stimulating isotype switching in B-cells. Furthermore, IL-27 has a significant impact on CD4 T-cell expansion, particularly naive cells.
IL-27 works synergistically with IL-12 to trigger the production of interferon-gamma (IFN-gamma) in naive CD4 T-cells. It binds to the cytokine receptor WSX-1/TCCR, which is necessary for IL-27-mediated signal transduction. Moreover, IL-27 potentiates the early phase of TH1 response while suppressing TH2 and TH17 differentiation.
The differentiation of TH1 cells is induced by IL-27 through two distinct pathways: the p38 MAPK/TBX21- and ICAM1/ITGAL/ERK-dependent pathways. It also leads to phosphorylation of STAT1, STAT3, STAT4, and STAT5, activating TBX21/T-Bet via STAT1. This activation results in the up-regulation of IL12RB2, which is critical for TH1 cell commitment. Additionally, IL-27 suppresses the expression of GATA3, an inhibitor of TH1 cell development.
In CD8 T-cells, IL-27 activates STATs and GZMB. Interestingly, IL-27 has a potent inhibitory effect on the development and production of IL-17 by TH17 cells. Even the IL-27 subunit p28 alone can inhibit IL-17 production by both CD4 and CD8 T-cells.
While IL-27 suppresses the development of pro-inflammatory TH17 cells through STAT1, it inhibits the development of anti-inflammatory regulatory T-cells (iTreg) independently of STAT1. Additionally, IL-27 has suppressive effects on pro-inflammatory cytokine production such as IL2, IL4, IL5, and IL6. It activates suppressors of cytokine signaling like SOCS1 and SOCS3. Furthermore, IL-27 can counteract the effects of certain cytokines, such as IL6, directly on T cells.
IL-27 also displays important antitumor and antiangiogenic activities. It activates the production of antiangiogenic chemokines like IP-10/CXCL10 and MIG/CXCL9. In vein endothelial cells, IL-27 induces IRF1/interferon regulatory factor 1 and up-regulates the expression of MHC class II transactivator/CIITA, resulting in the up-regulation of major histocompatibility complex class II. Additionally, IL-27 exhibits antiviral activity by inhibiting HIV-1 replication.

Associates with EBI3 to form the IL-27 interleukin, a heterodimeric cytokine which functions in innate immunity. IL-27 has pro- and anti-inflammatory properties, that can regulate T-helper cell development, suppress T-cell proliferation, stimulate cytotoxic T-cell activity, induce isotype switching in B-cells, and that has diverse effects on innate immune cells. Among its target cells are CD4 T-helper cells which can differentiate in type 1 effector cells (TH1), type 2 effector cells (TH2) and IL17 producing helper T-cells (TH17). It drives rapid clonal expansion of naive but not memory CD4 T-cells. It also strongly synergizes with IL-12 to trigger interferon-gamma/IFN-gamma production of naive CD4 T-cells, binds to the cytokine receptor WSX-1/TCCR which appears to be required but not sufficient for IL-27-mediated signal transduction.
