Description | The regulation of transcription in eukaryotes heavily relies on the modulation of chromatin structure. Post-translational modifications such as acetylation, phosphorylation, methylation, and ubiquitination of the amino-terminal tails of core histones play a critical role in modulating gene expression by altering chromatin accessibility to transcription factors (2-5). Typically, H2B is acetylated at specific lysine residues 5, 12, 15, and 20, while H3 is acetylated at lysine 9, 14, 18, 23, 27, and 56. Notably, acetylation at lysine 9 of H3 appears to play a dominant role in histone deposition and chromatin assembly in some organisms (2,3). Meanwhile, phosphorylation of H3 at serine 10, 28, and threonine 11 correlates tightly with chromosomal condensation during mitosis and meiosis (8-10), whereas threonine 3 of H3 is conserved among many species and is catalyzed by haspin kinase (11). Phospho-specific antibodies staining of mammalian cells reveals mitotic phosphorylation of H3 threonine 3 during prophase, followed by its dephosphorylation in anaphase (11). |
