Overview

Description

The FLI1 protein is a member of the ETS family and acts as a sequence-specific transcriptional activator. It recognizes the DNA sequence 5'-C[CA]GGAAGT-3'. Notably, it has the capability to form homodimers or heterodimers with ETV6/TEL1. Interestingly, alternative splicing gives rise to 2 isoforms of this protein in humans.

Other names

The ERGB transcription factor, also known as Ewing Sarcoma breakpoint region 2 (EWSR2), FLI 1 proto-oncogene, or FLI1, is a critical player in various biological processes. It has been implicated in the pathogenesis of Ewing Sarcoma, a highly malignant bone tumor. The fusion gene FLI1 EWS, resulting from the translocation between FLI1 and EWSR2 genes, enhances the oncogenic potential of FLI1.
FLI1 is a member of the ETS transcription factor family and is involved in regulating gene expression. It is known to play a significant role in leukemogenesis and tumor development. The Friend leukemia integration 1 (FLI1) transcription factor has been extensively studied for its contribution to the development of various cancers.
The FLI1 protein is encoded by the FLI1 gene and is also referred to as the Proto-oncogene Fli-1. It is essential for normal hematopoiesis and vascular development, but aberrant expression or fusion with other genes can lead to malignant transformation.
The Sarcoma breakpoint region 2 (EWSR2) is a region involved in chromosomal translocations with FLI1, leading to the formation of the FLI1 EWS fusion gene. This fusion gene is commonly observed in Ewing Sarcoma and contributes to the pathogenesis of this aggressive cancer.
The exact function of the Viral integration region FLI1, mouse, homolog of (also known as SIC1 or Viral integration region FLI1) is still unclear. However, it is believed to have a similar role to the human FLI1 gene in regulating gene expression and oncogenic processes.
Overall, the ERGB transcription factor, or FLI1, is a critical player in the development and progression of various cancers, including Ewing Sarcoma. Further research on its molecular mechanisms and its potential as a therapeutic target is warranted.

Specificity

The detection of total FLI1 at its endogenous levels can be accomplished using the FLI1 antibody. To generate a content that is highly similar but rearranged, it can be said that the FLI1 antibody is capable of recognizing and quantifying the native expression levels of FLI1 protein in a sample. By utilizing the FLI1 antibody, researchers can gain insights into the overall presence of FLI1 and its regulation in biological systems.

Application

The recommended dilutions for western blotting (WB) range from 1:500 to 1:3000. For immunohistochemistry (IHC), the dilutions typically range from 1:50 to 1:200. In the case of immunofluorescence/immunocytochemistry (IF/ICC), the suggested dilutions are usually between 1:100 and 1:500. For ELISA using peptides, the recommended dilutions are quite low, ranging from 1:20000 to 1:40000. Please note that these dilutions are based on the original information provided and should be used as a reference for experimentation.

Immunogen

A synthesized peptide derived from human FLI1.
