Description

This particular gene encodes a protein that belongs to the immunophilin protein family. Immunophilins play critical roles in immunoregulation as well as fundamental cellular processes that involve protein folding and transportation. The protein produced by this gene acts as a cis-trans prolyl isomerase and has the ability to bind to immunosuppressants such as FK506 and rapamycin.
Interestingly, this protein bears a striking resemblance in both structure and function to FK506-binding protein 1A (FKBP1A). However, unlike FKBP1A, this protein does not possess any immunosuppressant activity when combined with FK506. Instead, it interacts with interferon regulatory factor-4 and plays a crucial role in regulating gene expression related to the immune response in B and T lymphocytes.
Furthermore, this protein is known to form associations with phytanoyl-CoA alpha-hydroxylase. It also has the ability to interact with two heat shock proteins, namely hsp90 and hsp70. Thus, it is likely involved in the intracellular transport of hetero-oligomeric forms of steroid hormone receptors.
A noteworthy observation is that this protein demonstrates a strong correlation with adeno-associated virus type 2 vectors (AAV). This correlation results in a significant enhancement of AAV-mediated transgene expression in various human cell lines. Consequently, this encoded protein holds considerable importance for the optimal utilization of AAV vectors in human gene therapy.
It is worth mentioning that the human genome encompasses multiple non-transcribed pseudogenes that bear resemblance to this gene, adding to the complexity of its role and regulation.

FKBP4, also known as HBI, Hsp56, or p59, is a 59 kDa protein that functions as a peptidyl-prolyl cis-trans isomerase (PPIase) and a chaperone. FKBP59 and p52 are alternative names for FKBP52, which is also an immunophilin and a co-chaperone of Hsp90. These proteins assist in the folding and trafficking of client proteins, particularly steroid hormone receptors and kinases. FKBP4 and FKBP52 are widely expressed in many tissues, including the brain, heart, and immune cells, and have been implicated in various diseases, such as cancer, neurodegeneration, and immunological disorders. N-terminally processed isoforms of FKBP52 have been identified that have distinct functions. FKBP4 and FKBP52 are important drug targets for immunosuppression and cancer therapy, as well as potential biomarkers for disease diagnosis and prognosis.

Immunophilin protein with PPIase and co-chaperone activities. Component of steroid receptors heterocomplexes through interaction with heat-shock protein 90 (HSP90). May play a role in the intracellular trafficking of heterooligomeric forms of steroid hormone receptors between cytoplasm and nuclear compartments. The isomerase activity controls neuronal growth cones via regulation of TRPC1 channel opening. Acts also as a regulator of microtubule dynamics by inhibiting MAPT/TAU ability to promote microtubule assembly. May have a protective role against oxidative stress in mitochondria.
