Description

This gene encodes the gamma component of fibrinogen, a glycoprotein found in the bloodstream that consists of three pairs of distinct polypeptide chains. When there is damage to blood vessels, fibrinogen is cleaved by thrombin to create fibrin, which comprises the bulk of blood clots. In addition to clot formation, fibrinogen and fibrin cleavage products play a crucial role in cell adhesion and spreading, have vasoconstrictor and chemotactic properties, and act as growth factors for a variety of cell types. Disorders such as dysfibrinogenemia, hypofibrinogenemia, and thrombophilia can arise from mutations in this gene. Two distinct isoforms result from alternative splicing of the mRNA.

Other namesThe FIBG_HUMAN gene encodes for the fibrinogen gamma chain, which is also known as the fibrinogen gamma polypeptide or the fibrinogen gamma-b chain. This chain is a critical component of the fibrinogen molecule, which plays an essential role in blood clotting. The gamma chain is involved in the binding of fibrinogen to integrin receptors on platelet surfaces, enabling platelets to aggregate and form clots. Any mutations in this gene that affect the production or functionality of the fibrinogen gamma chain can lead to bleeding disorders or other blood-related conditions. Therefore, understanding the role of this chain in blood clotting is crucial for developing effective treatments for these disorders.

SpecificityThe FGG Antibody is capable of detecting the total amount of endogenous FGG present in a sample. This implies that the antibody can successfully identify and quantify all forms and variants of FGG that exist naturally in the sample.

AntigenFGG
ApplicationThe recommended dilution for Western Blotting is between 1:500 to 1:2000, while for Immunohistochemistry it ranges from 1:50 to 1:200. For ELISA with peptide, the optimal dilution is 1:20000-1:40000. These dilution ranges are crucial to get accurate and reliable results during the experiments. It is important to follow the recommended dilution and ensure that all the necessary precautions and techniques are considered to optimize the performance of the assays.

Target

Background

Together with fibrinogen alpha (FGA) and fibrinogen beta (FGB), polymerizes to form an insoluble fibrin matrix. Has a major function in hemostasis as one of the primary components of blood clots. In addition, functions during the early stages of wound repair to stabilize the lesion and guide cell migration during re-epithelialization. Was originally thought to be essential for platelet aggregation, based on in vitro studies using anticoagulated blood. However, subsequent studies have shown that it is not absolutely required for thrombus formation in vivo. Enhances expression of SELP in activated platelets via an ITGB3-dependent pathway. Maternal fibrinogen is essential for successful pregnancy. Fibrin deposition is also associated with infection, where it protects against IFNG-mediated hemorrhage. May also facilitate the antibacterial immune response via both innate and T-cell mediated pathways.

Tissue specificityDetected in blood plasma (at protein level).
Posttranslational modificationConversion of fibrinogen to fibrin is triggered by thrombin, which cleaves fibrinopeptides A and B from alpha and beta chains, and thus exposes the N-terminal polymerization sites responsible for the formation of the soft clot. The soft clot is converted into the hard clot by factor XIIIA which catalyzes the epsilon-(gamma-glutamyl)lysine cross-linking between gamma chains (stronger) and between alpha chains (weaker) of different monomers.Sulfation of C-terminal tyrosines increases affinity for thrombin.
Celluar localizationEndoplasmic reticulum;Extracellular region or secreted;Plasma Membrane;
