Description

Ephrin-B2 receptor, a crucial player in differentiation and development, specifically binds to members of the ephrin-B family. Its involvement in mediating various events is well-established.

Other namesThe human EphB4 gene codes for the protein called ephrin receptor EphB4, which belongs to the family of receptor tyrosine kinases. This protein is involved in cellular communication and plays a crucial role in various developmental processes. It is also known by other names, such as Hepatoma Transmembrane Kinase (HTK), MYK 1, and TYRO 11.
The expression of EphB4 is observed in various tissues, including the nervous system, heart, and intestines. It binds to the ephrin-B ligands, and upon binding, activates signaling pathways that regulate cellular processes like cell migration and adhesion. Moreover, EphB4 signaling is known to be involved in angiogenesis, the formation of new blood vessels from pre-existing ones, which is crucial for tumor growth.
Mutations in the EphB4 gene have been associated with various diseases, including cancer, cardiovascular disorders, and Alzheimer's disease. Therefore, understanding the role of the EphB4 receptor in normal and diseased states is of great therapeutic importance.

Target

Background

Receptor tyrosine kinase which binds promiscuously transmembrane ephrin-B family ligands residing on adjacent cells, leading to contact-dependent bidirectional signaling into neighboring cells. The signaling pathway downstream of the receptor is referred to as forward signaling while the signaling pathway downstream of the ephrin ligand is referred to as reverse signaling. Together with its cognate ligand/functional ligand EFNB2 plays a central role in heart morphogenesis and angiogenesis through regulation of cell adhesion and cell migration. EPHB4-mediated forward signaling controls cellular repulsion and segregation form EFNB2-expressing cells. Plays also a role in postnatal blood vessel remodeling, morphogenesis and permeability and is thus important in the context of tumor angiogenesis.

Tissue specificityAbundantly expressed in placenta but also detected in kidney, liver, lung, pancreas, skeletal muscle and heart. Expressed in primitive and myeloid, but not lymphoid, hematopoietic cells. Also observed in cell lines derived from liver, breast, colon, lung, melanocyte and cervix.
Posttranslational modificationPhosphorylated; autophosphorylation is stimulated by EFNB2.
Celluar localizationCytosol;Extracellular region or secreted;Plasma Membrane;
