It is sometimes referred to as AQPX2 or Aquaporin X2. Aquaporin-12A is one of the less-studied aquaporins, and its physiological functions are not yet fully understood. However, it is expressed in some tissues that are involved in fluid secretion or absorption, such as the salivary gland, kidney, and epididymis. The AQP12A gene is located on chromosome 2 in humans and contains 4 exons. The predicted amino acid sequence of human AQP12A consists of 329 residues and shares high sequence similarity with other aquaporin isoforms, especially AQP11 and AQP6. AQP12A has six transmembrane domains (TM1-TM6) connected by five loops (A-E) and two short terminal tails (N- and C-termini). The loops contain some conserved residues that are critical for water permeation and selectivity. Despite its name, AQP12A may also function as a glycerol channel in addition to a water channel. This is based on the fact that AQP12A can transport glycerol in vitro, and it has some structural features that are similar to known glycerol channels such as GlpF and AQP3. However, the physiological significance of glycerol transport by AQP12A is not clear yet. In conclusion, aquaporin 12A is a novel aquaporin with potential roles in fluid dynamics and possibly glycerol metabolism.
