Description

This mRNA-binding protein is a crucial player in the process of translation elongation. Its role extends beyond just protein synthesis, as it is also involved in regulating mRNA turnover downstream of decapping. Additionally, it contributes to maintaining cell wall integrity and responds to stress. It coordinates with syntenin SDCBP to regulate the activation of p53/TP53-dependent apoptosis, and is also involved in the regulation of TNF-alpha-mediated apoptosis. This protein has been shown to be crucial for extending and protecting neuronal processes, and in influencing brain development and function, as well as skeletal muscle stem cell differentiation. Moreover, it has been identified as a cofactor for functionally important viral proteins like HTLV-1 Rex and HIV-1 Rev that are essential for mRNA export of retroviral transcripts.

Other namesEukaryotic initiation factor 4D, also known as eIF-4D, is a protein involved in the regulation of protein synthesis. It helps in the initiation of protein translation by binding with the mRNA and ribosomes. On the other hand, eukaryotic initiation factor 5A, or eIF-5A, is involved in the elongation and termination phases of protein synthesis. It also has a role in cellular stress responses and is essential for cell growth and proliferation.
There are two isoforms of eukaryotic initiation factor 5A - eIF-5A-1 and eIF5A. These proteins are structurally similar and have overlapping functions. They are both involved in the regulation of gene expression and play a crucial role in cell survival and development.
Another name for eukaryotic initiation factor 5A is Rev-binding factor or uORF A. It is also known as EIF5A1 or IF5A1_HUMAN and has two alternative names MGC104255 and MGC99547.
Overall, these proteins play critical roles in protein synthesis, gene expression, and cellular processes. Understanding their functions is crucial in the development of targeted therapies for various diseases.

Target

Background

mRNA-binding protein involved in translation elongation. Has an important function at the level of mRNA turnover, probably acting downstream of decapping. Involved in actin dynamics and cell cycle progression, mRNA decay and probably in a pathway involved in stress response and maintenance of cell wall integrity. With syntenin SDCBP, functions as a regulator of p53/TP53 and p53/TP53-dependent apoptosis. Regulates also TNF-alpha-mediated apoptosis. Mediates effects of polyamines on neuronal process extension and survival. May play an important role in brain development and function, and in skeletal muscle stem cell differentiation. Also described as a cellular cofactor of human T-cell leukemia virus type I (HTLV-1) Rex protein and of human immunodeficiency virus type 1 (HIV-1) Rev protein, essential for mRNA export of retroviral transcripts.

Tissue specificityExpressed in umbilical vein endothelial cells and several cancer cell lines (at protein level).
Posttranslational modificationAcetylated. Deacetylated by SIRT2.eIF-5A seems to be the only eukaryotic protein to have a hypusine residue which is a post-translational modification of a lysine by the addition of a butylamino group (from spermidine).
Celluar localizationCytosol;Endoplasmic reticulum;Extracellular region or secreted;Nucleus;
UniPortP63241
