Description | Cbl proteins, including c-Cbl, Cbl-b, and Cbl-c, belong to the Casitas B lineage lymphoma family of single subunit RING finger protein-ubiquitin E3 ligases. The N-terminal tyrosine kinase-binding (TKB) domain of Cbl proteins enables them to interact with phosphorylated tyrosine residues on Cbl substrates, while the proline-rich domains, C-terminal to the RING finger, mediate interactions with SH3 domain-containing proteins. As adaptor proteins, Cbl proteins target specific substrates for ubiquitination and degradation, and they also play a role in receptor tyrosine kinase internalization and activation of signaling pathways. In particular, Cbl-b is a well-studied E3 ubiquitin ligase that downregulates TCR expression during antigen recognition and acts as a negative regulator of CD28 signaling cascade by ubiquitinating the p85 regulatory subunit of PI3K. Cbl-b mRNA and protein are upregulated in T cells following calcium mobilization and calcineurin activation, and its deficiency can lead to resistance to anergy induction due to defects in the ubiquitination of PLCγ1 and PKCθ. |

Background | E3 ubiquitin-protein ligase which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes, and transfers it to substrates, generally promoting their degradation by the proteasome. Negatively regulates TCR (T-cell receptor), BCR (B-cell receptor) and FCER1 (high affinity immunoglobulin epsilon receptor) signal transduction pathways.
