Description

Epigenetic modifications, such as cytosine methylation of DNA in mammalian cells, are heritable and play a critical role in gene expression regulation, development, and genomic imprinting. The mammalian DNA methyltransferase family is composed of three types: DNMT1, DNMT2, and DNMT3. DNMT1 is expressed in proliferating cells and acts as a maintenance methyltransferase during DNA replication. DNMT3A and DNMT3B are highly expressed in embryonic stem cells and function as de novo methyltransferases that methylate previously unmethylated regions of DNA. DNMT2, on the other hand, plays a minimal role in DNA methylation and is expressed at low levels in adult somatic tissues. Additionally, DNMT3L is a regulatory factor for DNMT3A and DNMT3B de novo methyltransferases that is primarily expressed in embryonic stem cells, testis, ovaries, and thymus. This factor consists of a heterotetrameric complex containing a methyltransferase-like domain that enhances the catalytic activity of DNMT3A and DNMT3B by increasing the binding of S-adenosylmethionine and DNA. Furthermore, the ADD domain helps the methyltransferase complex specifically target transcriptionally inactive regions of the genome by binding to unmethylated histone H3 Lys4.

Other namesPossible rearranged content:
- DNA cytosine 5 methyltransferase 3 like protein, also known as DNMT3L or DNA (cytosine-5)-methyltransferase 3-like, is a mammalian enzyme that plays a role in epigenetic regulation of gene expression.
- Like other DNA methyltransferases, DNMT3L catalyzes the transfer of a methyl group from S-adenosylmethionine to the C5 position of cytosine residues in DNA, leading to the formation of 5-methylcytosine (5mC).
- However, DNMT3L lacks the catalytic domain required for efficient methylation, and is therefore considered a catalytically inactive cofactor or regulator of other DNA methyltransferases, specifically DNMT3A and DNMT3B.
- DNMT3L interacts with these enzymes through its N-terminal domain, which contains a conserved PWWP domain that recognizes histone H3 lysine 36 trimethylation (H3K36me3), an epigenetic mark associated with actively transcribed genes and DNA replication.
- By binding to H3K36me3, DNMT3L can recruit DNMT3A and DNMT3B to chromatin regions that are enriched with this mark, and facilitate their establishment of de novo DNA methylation patterns during development, imprinting, and X chromosome inactivation.
- In addition to its role in methyltransferase activity, DNMT3L has been implicated in other cellular processes, such as RNA splicing, alternative polyadenylation, and retrotransposon silencing.
- Its expression is regulated by various factors, including DNA methylation, histone modifications, and transcription factors such as Oct4, Sox2, and Nanog, which are key regulators of pluripotency and differentiation in embryonic stem cells.
- Aberrant expression or mutations in DNMT3L have been associated with various diseases and disorders, such as cancer, neuropathies, and male infertility. Therefore, DNMT3L represents a potential target for diagnostic and therapeutic interventions in these conditions.

SpecificityTotal DNMT3L levels can be detected using the DNMT3L antibody, which specifically targets the endogenous levels of the protein. To generate a highly similar content, we can rephrase the information as follows: The detection of total DNMT3L levels can be achieved through the use of the DNMT3L antibody. This specific antibody is capable of identifying the endogenous levels of DNMT3L protein.

Target

Background

Catalytically inactive regulatory factor of DNA methyltransferases that can either promote or inhibit DNA methylation depending on the context (By similarity). Essential for the function of DNMT3A and DNMT3B: activates DNMT3A and DNMT3B by binding to their catalytic domain (PubMed:17687327). Acts by accelerating the binding of DNA and S-adenosyl-L-methionine (AdoMet) to the methyltransferases and dissociates from the complex after DNA binding to the methyltransferases (PubMed:17687327). Recognizes unmethylated histone H3 lysine 4 (H3K4me0) and induces de novo DNA methylation by recruitment or activation of DNMT3 (PubMed:17687327). Plays a key role in embryonic stem cells and germ cells (By similarity). In germ cells, required for the methylation of imprinted loci together with DNMT3A (By similarity). In male germ cells, specifically required to methylate retrotransposons, preventing their mobilization (By similarity). Plays a key role in embryonic stem cells (ESCs) by acting both as an positive and negative regulator of DNA methylation (By similarity). While it promotes DNA methylation of housekeeping genes together with DNMT3A and DNMT3B, it also acts as an inhibitor of DNA methylation at the promoter of bivalent genes (By similarity). Interacts with the EZH2 component of the PRC2/EED-EZH2 complex, preventing interaction of DNMT3A and DNMT3B with the PRC2/EED-EZH2 complex, leading to maintain low methylation levels at the promoters of bivalent genes (By similarity). Promotes differentiation of ESCs into primordial germ cells by inhibiting DNA methylation at the promoter of RHOX5, thereby activating its expression (By similarity).
