Description | CYP1B1 is a crucial enzyme found in normal breast tissue and breast cancer, responsible for producing potentially cancer-causing estrogen metabolites and activating environmental carcinogens. It is a dominant member of the CYP1 family. Breast cancer development is believed to be triggered by estrogen through the formation of its metabolite, catechol estrogen (CE). The conversion of 17-beta-estradiol to catechol estrogen metabolites 2-OH-E2 and 4-OH-E2, catalyzed by CYP1B1, has been associated with mammary carcinogenesis. Additionally, genetic variations in CYP1B1 may also play a significant role in prostate carcinogenesis. A specific polymorphism at codon 432 of the CYP1B1 gene (Val->Leu) is linked to alterations in its catalytic function. |

Background | Cytochromes P450 are a group of heme-thiolate monooxygenases. In liver microsomes, this enzyme is involved in an NADPH-dependent electron transport pathway. It oxidizes a variety of structurally unrelated compounds, including steroids, fatty acids, retinoid and xenobiotics. Preferentially oxidizes 17beta-estradiol to the carcinogenic 4-hydroxy derivative, and a variety of procarcinogenic compounds to their activated forms, including polycyclic aromatic hydrocarbons. Promotes angiogenesis by removing cellular oxygenation products, thereby decreasing oxidative stress, release of antiangiogenic factor THBS2, then allowing endothelial cells migration, cell adhesion and capillary morphogenesis.
