Description

Myosin is a protein complex made up of six polypeptide chains. The complex features two heavy chains that are identical, and two pairs of light chains. Myosin light chain 2 or MLC2 is also known as myosin regulatory light chain (MRLC), RLC, or LC20, depending on its distribution and the type of muscle it is present in.
In smooth muscle, MLC2 is phosphorylated at Thr18 and Ser19 by myosin light chain kinase (MLCK) in a Ca2+/calmodulin-dependent manner. This phosphorylation has been linked to myosin ATPase activity and smooth muscle contraction. When Ser19 of smooth muscle MLC2 is phosphorylated by ROCK, it regulates the assembly of stress fibers. PKC and cdc2 have been shown to phosphorylate Ser1/Ser2 and Ser9 of smooth muscle MLC2, respectively, causing the inhibition of myosin ATPase activity. CDC2's role in phosphorylating MLC2 also is responsible for controlling the timing of cytokinesis.
Transgenic mice without phosphorylation sites on the cardiac muscle isoform exhibited morphological and functional abnormalities which illustrate the importance of phosphorylation for proper muscle function. In summary, phosphorylation plays a crucial role in the regulation of myosin ATPase activity, muscle contraction, and cytokinesis, making it an essential mechanism in muscle function.
