Background

This enzyme plays a crucial role in the transsulfuration pathway, serving as the catalyst for the final step in the conversion of methionine to cysteine. It displays a wide range of substrate acceptance, exhibiting broad specificity. Its primary function is to transform cystathionine into cysteine, ammonia, and 2-oxobutanoate. Interestingly, it also has the capability to convert two cysteine molecules into lanthionine and hydrogen sulfide. Moreover, it can accept homocysteine as a substrate, although its specificity depends on the levels of the endogenous substrates present. One of its key contributions lies in its ability to generate hydrogen sulfide (H2S), an important signaling molecule involved in the regulation of blood pressure.
Defects in CTH are the cause of cystathioninuria (CSTNU). It is an autosomal recessive phenotype characterized by abnormal accumulation of plasma cystathionine, leading to increased urinary excretion.
