Overview

Description

CBP is a protein acetyltransferase that plays a critical role in transcriptional activation of histones. It has been reported to acetylate NCOA3 coactivator and bind to phosphorylated CREB1, enhancing its transcriptional activity towards cAMP-responsive genes. However, methylation of the KIX domain by CARM1 has been found to block CBP's association with CREB, resulting in CREB signaling blockade and an activated apoptotic response.
CBP is typically found in a complex containing NCOA2, NCOA3, IKKA, IKKB, and IKBKG, and is believed to be part of a complex with HIF1A and EP300. It has also been shown to interact with the C-terminal region of CITED4, while the TAZ-type 1 domain interacts with HIF1A. Other molecules that interact with CBP include MAF, SRCAP, CARM1, ELF3, MLLT7/FOXO4, N4BP2, NCOA1, NCOA3, NCOA6, PCAF, PELP1, PML, SMAD1, SMAD2, SMAD3, SPIB and TRERF1.
Besides these interactions, CBP has been found to interact with HTLV-1 Tax, p30II, and HIV-1 Tat, KLF1, and more. These findings suggest that CBP has widespread effects on gene transcription and may play a crucial role in regulating various physiological and pathological processes.

Other names

The human CBP gene, also known as CREB-binding protein, is responsible for encoding a protein that plays a crucial role in regulating gene expression. CBP, or CREB binding protein, is also referred to as Crebbp, KAT3A, or cyclic AMP responsive enhancer binding protein. Mutations in this gene have been linked to Rubinstein-Taybi syndrome (RTS), a genetic disorder characterized by intellectual disability, distinctive facial features, and broad thumbs and toes. Overall, CBP is a vital player in the mechanism of gene transcription and its dysregulation can lead to serious health consequences.

Application

The suggested dilution ranges for WB are 1:500-1:2000, for IHC are 1:50-1:200, for IF/ICC are 1:100-1:500, and for ELISA(peptide) are 1:20000-1:40000. To rearrange the content and maintain its original information, the following revised version is provided:
For Western Blot (WB), it is recommended to use a dilution range of 1:500-1:2000. This range ensures optimal detection of the target protein.
Immunohistochemistry (IHC) experiments should consider a dilution range of 1:50-1:200. Diluting the primary antibody within this range will provide accurate staining and reliable results.
For Immunofluorescence (IF) or Immunocytochemistry (ICC), it is advisable to dilute the primary antibody in a range of 1:100-1:500. This dilution range enables the efficient labeling of target proteins within the samples.
When performing ELISA assays with peptide antigens, it is recommended to dilute the primary antibody in a range of 1:20000-1:40000. This dilution range ensures precise and sensitive detection of the peptide in the samples.
Please note that these dilution ranges serve as general guidelines. Optimization may be required for specific experiments and antibody concentrations can vary between different antibodies.

Stability & Storage

To preserve the quality of the product, it is recommended to store it at a temperature of -20 °C for a duration of one year. It is important to avoid subjecting the product to repeated cycles of freezing and thawing as this may affect its efficacy and potency. Hence, it is advisable to take appropriate precautions while storing the product to ensure its longevity and effectiveness.

Storage buffer

This solution contains Rabbit IgG in a phosphate buffered saline solution with a pH of 7.4, 150mM NaCl and 0.02% sodium azide, along with 50% glycerol. It should be stored at a temperature of -20°C and is stable for a period of 12 months from the date of receipt. This ensures that the product remains in good condition for use over an extended period, allowing for efficient and reliable results in any laboratory setting.

Target

Background

Histone acetylation is a process that adds an acetyl group to histones, resulting in a specific tag for activating gene transcription. This modification not only affects histones but also extends to non-histone proteins such as NCOA3 and FOXO1. It binds specifically to phosphorylated CREB, boosting its ability to activate genes responsive to cAMP. Furthermore, it acts as a coactivator for ALX1 and enhances the activity of circadian transcriptional activators NPAS2-ARNTL/BMAL1 and CLOCK-ARNTL/BMAL1 heterodimers. Another important role is acetylating PCNA, which promotes the elimination of chromatin-bound PCNA and its degradation during nucleotide excision repair (NER) (PubMed:24939902). By rearranging the provided information, the following content maintains its essence while being expressed differently.

Posttranslational modification

Methylation of the KIX domain by CARM1 blocks association with CREB. This results in the blockade of CREB signaling, and in activation of apoptotic response (By similarity).Phosphorylated by CHUK/IKKA at Ser-1382 and Ser-1386; these phosphorylations promote cell growth by switching the binding preference of CREBBP from TP53 to NF-kappa-B.Sumoylation negatively regulates transcriptional activity via the recruitment of DAAX.Autoacetylation is required for binding to protein substrates, such as acetylated histones and acetylated TP53/p53.
