Description

BID, a member of the Bcl-2/Bcl-xL family, is a death agonist protein that contains the BH3 domain. Originally, it localizes in the cytosolic fraction of cells as an inactive precursor. However, upon cleavage by caspase-8 in the Fas signaling pathway, BID is activated and translocates to mitochondria, inducing mitochondrial damage and cytochrome c release. As a result, BID plays a significant role in transmitting apoptotic signals from the cell surface to mitochondria. Nonetheless, the exact mechanism by which cleaved BID translocates and cytochrome c is released remains unclear.

Other namesBID, also known as apoptotic death agonist or BH3 interacting domain death agonist, is a protein involved in the apoptotic pathway. It has various isoforms, including BID isoform ES(1b), BID isoform L(2), and BID isoform Si6. Other related terms for BID include BH3 interacting domain death agonist p11, BH3 interacting domain death agonist p13, BH3 interacting domain death agonist p15, and BH3-interacting domain death agonist p11. Additionally, BID is also referred to as p11 BID, p13 BID, p15 BID, or p22 BID. Another name for BID is FP497. It is encoded by the human BID coding sequence and is also known as MGC15319 or MGC42355. Along with its association with apoptosis, BID is sometimes linked to desmocollin type 4.

AntigenBID
ApplicationThe recommended dilution range for WB is from 1:500 to 1:2000, while for IHC it is 1:50 to 1:200. For ELISA using peptide, it is suggested to use a dilution of 1:20000 to 1:40000. These dilution ranges are based on optimal results from previous tests and are intended to ensure accurate and reliable detection of the target protein. It is important to follow the instructions carefully and adjust the dilution ratio as needed to achieve the best possible results for your specific application.

Target

Background

The major proteolytic product p15 BID allows the release of cytochrome c (By similarity). Isoform 1, isoform 2 and isoform 4 induce ICE-like proteases and apoptosis. Isoform 3 does not induce apoptosis. Counters the protective effect of Bcl-2.

Tissue specificityIsoform 2 and isoform 3 are expressed in spleen, bone marrow, cerebral and cerebellar cortex. Isoform 2 is expressed in spleen, pancreas and placenta (at protein level). Isoform 3 is expressed in lung, pancreas and spleen (at protein level). Isoform 4 is expressed in lung and pancreas (at protein level).
Posttranslational modificationTNF-alpha induces a caspase-mediated cleavage of p22 BID into a major p15 and minor p13 and p11 products.p15 BID is ubiquitinated by ITCH; ubiquitination results in proteasome-dependent degradation.
Celluar localizationCytosol;Extracellular region or secreted;Mitochondrion;
