Background | This particular gene has been identified as a frequent target for mutations in various cancers, making it a crucial tumor suppressor. The encoded protein serves as a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase and possesses both a tensin-like domain and a catalytic domain similar to dual specificity protein tyrosine phosphatases. Unlike typical protein tyrosine phosphatases, this protein exhibits a preference for dephosphorylating phosphoinositide substrates. Its primary function involves the negative regulation of phosphatidylinositol-3,4,5-trisphosphate levels within cells, thereby acting as a tumor suppressor by exerting a negative influence on the AKT/PKB signaling pathway. Interestingly, an alternative translation start site, marked by a non-canonical (CUG) codon, generates a longer isoform that commences with a leucine amino acid. This isoform is believed to have a specific association with the inner membrane of mitochondria and potentially contributes to the regulation of energy metabolism within these organelles. Additionally, a pseudogene of this gene can be found on chromosome 9. Due to alternative splicing and the utilization of multiple translation start codons, different isoforms are produced, resulting in various transcript variants. |
