Overview

Other names

MAP Kinase Kinase 1, also known as MEK1 or MKK1, is a dual specificity enzyme that plays a crucial role in the MAPK/ERK signaling pathway. This enzyme phosphorylates and activates ERK1/2, leading to the regulation of various cellular processes such as proliferation, differentiation, and survival. It is also involved in a variety of pathological conditions, including cancer, neurodegenerative diseases, and cardiovascular disorders. MEK1 is highly conserved across species and is a promising target for therapeutic interventions. Further research on the regulation and function of this enzyme can provide valuable insights into the pathogenesis of several diseases and the development of new drugs.

Specificity

The detection of endogenous levels of MEK1 is facilitated by the Phospho-MEK1 (Thr291) Antibody, specifically when MEK1 is phosphorylated at Threonine 291. To generate similar content by rearranging the original text's information: The Phospho-MEK1 (Thr291) Antibody is capable of detecting the presence of phosphorylated MEK1 at Threonine 291, allowing for the measurement of endogenous levels of this protein.

Stability & Storage

In order to preserve the quality of the product, it is recommended to store it at a temperature of -20 °C for up to one year. It is important to avoid exposing the product to repeated freeze/thaw cycles, as this can have a detrimental effect on its stability and efficacy. By following these storage guidelines, you can ensure that the product remains in optimal condition for use when needed.

Storage buffer

The storage condition for Rabbit IgG is at -20 °C in phosphate buffered saline with a pH of 7.4, containing 150mM NaCl and 0.02% sodium azide. Additionally, it is supplemented with 50% glycerol. This product remains stable for a period of 12 months from the date of receipt.

Target

Background

As a dual specificity protein kinase, this protein plays a critical role in the MAP kinase signal transduction pathway. Upon binding of extracellular ligands such as growth factors, cytokines and hormones to their respective cell-surface receptors, the RAS pathway is activated, which leads to RAF1 activation. The latter in turn triggers the activation of the dual-specificity protein kinases MAP2K1/MEK1 and MAP2K2/MEK2. These kinases are specific to the MAPK/ERK cascade and catalyze the simultaneous phosphorylation of a threonine and tyrosine residue within the Thr-Glu-Tyr sequence present in the extracellular signal-regulated kinase MAPK3/ERK1 and MAPK1/ERK2. This results in the activation of these kinases, leading to further signal transduction within the MAPK/ERK cascade.
The MAPK/ERK pathway plays a crucial role in various biological processes such as cell growth, adhesion, survival, and differentiation. These effects are mainly mediated by the regulation of transcription, metabolism, and cytoskeletal rearrangements. This pathway also regulates endosomal dynamics, lysosome processing, endosome cycling through the perinuclear recycling compartment (PNRC), and Golgi fragmentation during mitosis.
One of the targets of the MAPK/ERK pathway is the peroxisome proliferator-activated receptor gamma (PPARG), a nuclear receptor that promotes differentiation and apoptosis. MAP2K1/MEK1 has been demonstrated to facilitate the export of PPARG from the nucleus. In conclusion, this dual-specificity protein kinase plays a major role in MAPK/ERK signaling and various cellular processes, and its dysregulation may contribute to the development of numerous diseases.

Tissue specificity

Widely expressed, with extremely low levels in brain.

Posttranslational modificationPhosphorylation at Ser-218 and Ser-222 by MAP kinase kinase kinases (RAF or MEKK1) positively regulates kinase activity. Also phosphorylated at Thr-292 by MAPK1/ERK2 and at Ser-298 by PAK. MAPK1/ERK2 phosphorylation of Thr-292 occurs in response to cellular adhesion and leads to inhibition of Ser-298 phosphorylation by PAK.Acetylation by Yersinia yopJ prevents phosphorylation and activation, thus blocking the MAPK signaling pathway.
