The NFKB complex is formed by the binding of either NFKB1 (MIM 164011) or NFKB2 (MIM 164012) with REL (MIM 164910), RELA (MIM 164014), or RELB (MIM 604758). In order to keep NF-kappa-B inactive and confined to the cytoplasm, I-kappa-B proteins (NFKBIA or NFKBIB, MIM 604495) play a crucial role by inhibiting the NFKB complex. These I-kappa-B proteins trap NF-kappa-B, preventing its activation. Hence, the balance between NFKB1/NFKB2 and I-kappa-B proteins is vital for regulating the activity of NF-kappa-B.
