Overview

Description

The receptor tyrosine kinase family known as the epidermal growth factor (EGF) receptor includes a specific gene coding for a protein. However, this protein doesn't have its own domain for binding growth factors, thus making it unable to bind to these factors effectively.

Other names

ErbB2, also known as HER2 or NEU, is a viral oncogene homolog that is associated with several types of cancers, including neuroblastoma and glioblastoma. It is a receptor tyrosine kinase that plays a crucial role in cellular growth and proliferation. The protein is encoded by the ERBB2 gene and is also referred to as CD340 or MLN19. Its overexpression is often observed in metastatic lymph nodes and is implicated in the progression of these cancers. ErbB2 is a target for therapeutic intervention and is known to be inhibited by herstatin. The avian homolog of this oncogene, V erb b2, is also implicated in erythroblastic leukemia.

The dilution ratios for Western Blotting (WB) range from 1:500 to 1:2000. For Immunohistochemistry (IHC), the suggested dilution ratios are 1:50 to 1:500. As for Immunofluorescence/Immunocytochemistry (IF/ICC), the appropriate dilution ratios are 1:100 to 1:500. Lastly, for Enzyme-linked Immunosorbent Assay (ELISA) using peptide antigens, the recommended dilution ratios are 1:20000 to 1:40000.

 Protein tyrosine kinase that is part of several cell surface receptor complexes, but that apparently needs a coreceptor for ligand binding. Essential component of a neuregulin-receptor complex, although neuregulins do not interact with it alone. GP30 is a potential ligand for this receptor. Regulates outgrowth and stabilization of peripheral microtubules (MTs). Upon ERBB2 activation, the MEMO1-RHOA-DIAPH1 signaling pathway elicits the phosphorylation and thus the inhibition of GSK3B at cell membrane. This prevents the phosphorylation of APC and CLASP2, allowing its association with the cell membrane. In turn, membrane-bound APC allows the localization of MACF1 to the cell membrane, which is required for microtubule capture and stabilization.

Posttranslational modificationAutophosphorylated. Autophosphorylation occurs in trans, i.e. one subunit of the dimeric receptor phosphorylates tyrosine residues on the other subunit (Probable). Ligand-binding increases phosphorylation on tyrosine residues (PubMed:27134172). Signaling via SEMA4C promotes phosphorylation at Tyr-1248 (PubMed:17554007).
