Background

The initiation of protein synthesis is controlled by EIF2, an important translation initiation factor. Its main function is to facilitate the binding of the initiator tRNA to the 40S ribosomal subunits, which is the first step in protein synthesis. This binding occurs in the form of a ternary complex composed of methionyl-tRNA, EIF2, and GTP. EIF2 itself is made up of three subunits: the 36-kD EIF2-alpha subunit (EIF2S1), the 38-kD EIF2-beta subunit (EIF2S2; MIM 603908), and the 52-kD EIF2-gamma subunit (EIF2S3; MIM 300161). The rate at which the ternary complex is formed is regulated by the phosphorylation status of the EIF2-alpha subunit. This important aspect was discovered by Ernst et al. in 1987 (PubMed 2948954). In summary, EIF2 plays a crucial role in the regulation of protein synthesis initiation through its interaction with the initiator tRNA and ribosomal subunits, and this process is influenced by the phosphorylation state of the EIF2-alpha subunit.
