Overview

Description

The recombinant PD1 protein aa1-165, which carried a C-terminal His tag, was successfully synthesized in mammalian cells. Its theoretical molecular weight was predicted to be 17.5 kDa. Subsequent purification led to the identification of the purified PD1 protein on an SDS-PAGE gel. Interestingly, under reduced conditions, it migrated near the position of 34 kDa. This observation suggests that the purified PD1 protein may undergo post-translational modifications or form stable dimers on the gel. The significant difference between the observed molecular weight and the predicted size warrants further investigations to elucidate the exact nature of this phenomenon. Nonetheless, this discovery provides valuable insights into the structural and functional properties of recombinant PD1 protein.

Proprietary chromatographic techniques are employed to purify PD1 protein. A highly similar version can be generated by rephrasing the sentence structure: "The purification of PD1 protein is carried out using exclusive chromatographic methods."

Entrez Gene summary for PD1:

This immunoglobulin superfamily gene encodes a membrane protein found on the cell surface. Its expression is predominantly observed in pro-B-cells, where it is believed to play a pivotal role in their differentiation process. In mice, when anti-CD3 antibodies are administered, it triggers the activation of this gene in the thymus, leading to a significant increase in apoptotic thymocytes.

UniProtKB/Swiss-Prot: Q15116 (PD1_HUMAN)

This specific cell surface receptor plays a crucial role in modulating T-cell activity during both immune responses and tolerance mechanisms. Its primary function is to act as an inhibitory molecule on T-cells upon binding to its ligand. By doing so, it selectively hinders the effector functions of T-cells in a manner that is specific to the targeted antigen. Furthermore, it has been suggested that in conjunction with other factors, this receptor might have the potential to induce cell death.
