Target

Background

Endoglin, a single-pass type I membrane protein, is primarily expressed on endothelial cells in all tissues except for bone marrow. It acts as a major glycoprotein in the vascular endothelium and can also be found on activated macrophages, fibroblasts, and smooth muscle cells. In addition, it forms a homodimer and can create a heteromeric complex with the signaling receptors TGFBR1 and/or TGFBR2, which are associated with transforming growth factor-beta. This complex might play a crucial role in the interaction between endothelial cells and integrins or other RGD receptors. Defects in the ENG gene are responsible for the development of hereditary hemorrhagic telangiectasia type 1 (HHT1), an autosomal dominant multisystemic vascular dysplasia. HHT1 is characterized by recurring nosebleeds (epistaxis), telangiectases on the skin and mucous membranes, gastrointestinal bleeding, and the formation of arteriovenous malformations in the pulmonary (PAVM), cerebral (CAVM), and hepatic systems.
