| Description | The BCA method is considered an excellent technique for quantifying proteins. It operates by forming complexes between the peptide bond structure of protein molecules and Cu2+ ions in an alkaline environment. This interaction reduces Cu2+ to Cu1+. A specific reagent called BCA binds specifically to Cu1+ and creates a stable purplish-blue complex. This complex exhibits maximum light absorption at 562nm, and the intensity of the color is directly proportional to the protein concentration. By measuring the absorption value, the protein concentration can be determined accurately. The BCA method offers numerous advantages, including its high sensitivity, simple procedure, and the stability of the reagent and color complex formed. It is also minimally affected by interfering substances like scale remover. The linear range for protein determination using BCA reagent is wide, ranging from 20 to 2000µg/mL. Compared to the classic Lowry method, the BCA method is four times faster, providing results within 45 minutes. Furthermore, the BCA method is not influenced by ionic and non-ionic detergents present in the samples, making it a versatile choice. Additionally, the coefficient of variation of different protein molecules in this method is much lower than that of Coomassie Brilliant Blue. Protein concentration can be determined using either the enzyme-plate method or spectrophotometer method. The composition of the BCA kit includes two forms of BCA protein quantitative liquid: A and B, which are available in volumes of 100mL and 125mL respectively for the 500 times concentration, and 4mL and 10mL respectively for the 2500 times concentration.
