Testing Principle：

The double antibody sandwich ELISA is used in this diagnostic kit, with a specific anti-human CXCL13 Capture Antibody pre-packaged on a high affinity conjugate plate. The standard sample and biotin labeled detection antibody are added one by one into the HRP-plate well. The sample is then incubated for 2 hours at room temperature, during which the CXCL13 combines with solid phase antibody and detection antibody. After thoroughly washing away unbound ingredients, horseradish peroxidase labeled streptavidin-HRP is introduced, followed by further washing. The addition of TMB color substrate triggers a color development process at room temperature in the dark, with the depth of the reaction positively correlated to the concentration of CXCL13 in the sample. The termination solution finally stops the reaction, and absorbance values are measured at a detection wavelength of 450 nm (with correction wavelength 570-630 nm) using a microplate analyzer.

Background：

IgE, also known as Immunoglobulin E, is an exclusive class of antibodies that can only be found in mammals. Its primary function revolves around providing immunity against parasites like worms, schistosomiasis mansoni, trichinella, and fluke hepatica. Remarkably, IgE plays a pivotal role in type I hypersensitivity reactions, including conditions like allergic asthma, most sinusitis cases, allergic rhinitis, food allergies, chronic urticaria, and atopic dermatitis. Moreover, it assumes significance in response to allergens, such as antigens utilized in allergic drugs, bee stings, and desensitization immunotherapy. Despite its minimal presence, accounting for just 0.05% of all Ig levels in normal human serum, IgE manages to instigate the most vigorous inflammatory response. Individuals with hereditary allergies may exhibit IgE levels up to 10 times higher in their bloodstream.
