To analyze the levels of SMPD1 protein in various cell lines, Western blot was performed using the SMPD1 antibody (abs117402) at a dilution of 1:1000. The secondary antibody used was the HRP Goat Anti-Rabbit IgG (H+L), which was diluted at 1:10000. For each lane, 25ug of lysates/proteins were used and blocked using 3% nonfat dry milk in TBST. The detection method used was the ECL Basic Kit, and the exposure time was set to 90 seconds. This procedure allowed for the detection and quantification of SMPD1 protein levels in the different cell lines.
