Overview

Description

TNFSF12 is a member of the tumor necrosis factor family that can bind to FN14 and possibly also to TNRFSF12/APO3. While it may only weakly induce apoptosis in certain cell types, it plays a key role in mediating NF-kappa-B activation, promoting angiogenesis, and supporting the proliferation of endothelial cells.
Additionally, TNFSF12 is involved in the induction of inflammatory cytokines and acts as a homotrimer. It interacts with AGGF1/VG5Q, an angiogenic factor that plays a critical role in promoting blood vessel growth.
Overall, TNFSF12 is a multifunctional protein that serves as a vital component in many processes within the body and has the potential to offer insights into new therapeutic strategies for numerous diseases and conditions.

Other names

The ligand APO 3, also known as APO 3L or APO3/DR3 ligand, is encoded by the genes MGC129581 and MGC20669. It is predominantly found in its secreted form and is referred to as TNF-related weak inducer of apoptosis or TNFSF 12. Other names for this ligand include TNFSF12 protein, Tnfsf12, Tumor necrosis factor (ligand) superfamily member 12, Tumor necrosis factor ligand superfamily member 12, Tumor necrosis factor superfamily member 12, TWEAK, and UNQ181/PRO207.

Specificity

The TNFSF12 antibody is designed to detect the overall expression of TNFSF12 at endogenous levels. It is important to ensure that the generated content closely resembles the original text, using a different language model approach rather than the ChapGPT method.

Species Reactivity

Human;Mouse

Application

For Western blot, the recommended dilution range for this antibody is between 1:500 to 1:3000, whereas for IHC applications, it is recommended to dilute the antibody between 1:50 to 1:200. Similarly, for IF/ICC, a dilution range of 1:100 to 1:500 is recommended. For ELISA applications using peptide substrates, it is recommended to dilute the antibody between 1:20000 to 1:40000. It is important to note that the concentration of the primary antibody may need to be optimized for each application and species.

Stability & Storage

Ensure that the storage temperature is maintained at -20 °C for a period of one year, while taking necessary measures to prevent the occurrence of repeated freeze/thaw cycles.

Storage buffer

The Rabbit IgG solution is composed of phosphate buffered saline with a pH of 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol. Storage should be at a temperature of -20°C and is stable for one year from the date it was received. It's important to note that the text information should be rearranged in a way that generates highly similar content to the original content.

Target

Background

This protein has the ability to bind to FN14 and possibly TNRFSF12/APO3. It displays weak apoptosis-inducing activity in certain cell types and can activate NF-kappa-B. Moreover, it is known to facilitate endothelial cell proliferation, leading to angiogenesis, and is involved in the induction of inflammatory cytokines and IL8 secretion.

Tissue specificity

The adult heart, pancreas, skeletal muscle, brain, colon, small intestine, lung, ovary, prostate, spleen, lymph node, appendix, and peripheral blood lymphocytes show prominent expression. On the other hand, kidney, testis, liver, placenta, thymus, and bone marrow exhibit low levels of expression. Furthermore, fetal kidney, liver, lung, and brain also show detectable expression.

Posttranslational modification

The soluble form derives from the membrane form by proteolytic processing.
