Description

The TNFR family comprises of 10 cell-surface proteins that play a crucial role in regulating the development and immune system of mammals. TNFRSF14, also known as TR2, ATAR, HVEA, HVEM, LIGHTR, is an important member of TNFR, which has been identified as a mediator of herpesvirus entry into mammalian cells. The cytoplasmic region of TNFRSF14 interacts with various members of the TRAF family, such as TRAF1, TRAF2, TRAF3, and TRAF5. When introduced into human 293 cells through transient transfection, TNFRSF14 led to significant activation of NF- B, which acts as a transcriptional regulator of numerous immunomodulatory and inflammatory genes.

Other namesThe Herpes virus entry mediator (HVEM) is a protein that plays a critical role in the immune response. HVEM, also known as CD270 or TNFRSF14, belongs to the tumor necrosis factor receptor superfamily. It is a cell surface receptor that binds to a variety of ligands, including the herpesvirus entry mediator ligand (HveA), LIGHT, and CD40 like protein precursor.
HVEM is expressed in a variety of immune cells, including T cells, B cells, and natural killer cells. Its main function is to regulate immune cell activation and survival. HVEM signaling can either promote or inhibit immune responses depending on the ligand that binds to it.
One of the most well-known functions of HVEM is its role in herpesvirus entry. Herpesviruses, such as herpes simplex virus (HSV), use HVEM to enter host cells. The virus binds to HVEM on the cell surface, which triggers a signaling cascade that allows the virus to enter the cell.
In addition to its role in viral entry, HVEM also plays a critical role in regulating immune cell function. Binding of LIGHT or CD40 ligand to HVEM can enhance immune responses, while binding of BTLA (a ligand that inhibits T cell activation) can suppress immune responses.
In summary, HVEM is a key player in both viral entry and immune regulation. Its diverse functions make it a promising target for therapeutic intervention in various diseases, including cancer and infectious diseases.

ApplicationFor Western Blotting, a recommended dilution range is between 1:500 and 1:2000. When performing Immunohistochemistry, it is suggested to use a dilution range of 1:50 to 1:200. As for ELISA using a peptide, a dilution range of 1:20000 to 1:40000 is recommended for optimal results. It is important to carefully follow these recommended dilution guidelines for each application to achieve accurate and reliable results.

Target

Background

Receptor for BTLA. Receptor for TNFSF14/LIGHT and homotrimeric TNFSF1/lymphotoxin-alpha. Involved in lymphocyte activation. Plays an important role in HSV pathogenesis because it enhanced the entry of several wild-type HSV strains of both serotypes into CHO cells, and mediated HSV entry into activated human T-cells.

Tissue specificityWidely expressed, with the highest expression in lung, spleen and thymus.
Posttranslational modificationN-glycosylated.
Celluar localizationPlasma Membrane;
