Description | The protein elongin B, encoded by this gene, is a crucial subunit of the transcription factor B (SIII) complex. Together with elongins A/A2 and C, it activates RNA polymerase II elongation by suppressing any transient pausing of the polymerase occurring within transcription units. While elongin A is the component of the SIII complex that actually triggers transcription, elongins B and C are regulatory subunits. In particular, elongin A2 is selectively expressed in the testis and is capable of forming a stable complex with elongins B and C. Interestingly, the von Hippel-Lindau tumor suppressor protein can interact with elongins B and C, and thus effectively inhibit transcription elongation. It should be noted that this gene has two alternatively spliced transcript variants that encode different isoforms. Additionally, pseudogenes have been discerned on chromosomes 11 and 13. |
| Other names | The protein EloB, also known as Elongin 18 kDa subunit or Elongin-B, is an important component of the RNA polymerase II transcription factor SIII. Its p18 subunit plays a key role in regulating transcription elongation. Additionally, it is also referred to as SIII p18 or TCEB 2 and is a crucial component of Transcription elongation factor B (SIII) polypeptide 2. Its function is to stimulate and improve elongation by RNA polymerase II, making it an essential transcription factor. The importance of this protein can be gauged by the fact that deficiencies in EloB have been linked to various developmental disorders and diseases. Therefore, its analysis and study holds great significance in unraveling the mysteries of transcriptional regulation. |

| Specificity | Total TCEB2 levels can be detected by the TCEB2 Antibody, which specifically targets the endogenous levels of this protein. To emphasize, the antibody is designed to identify the overall presence of TCEB2 within a biological sample. By using this antibody, researchers are able to accurately measure the total amount of TCEB2 present, providing valuable insights into its expression and potential functions. |

| Application | The recommended dilutions for using this antibody are 1:500-1:2000 for western blot and 1:50-1:200 for immunohistochemistry. When using this product for ELISA with peptide, a dilution of 1:20000-1:40000 is recommended. These concentrations have been found to be effective in detecting the target protein and producing reliable results. It is important to properly dilute the antibody before use in order to achieve optimal performance. With careful handling and proper storage, this antibody can provide highly specific and sensitive detection of the target antigen in a variety of experimental settings. |

Background | SIII, also known as elongin, is a general transcription elongation factor that increases the RNA polymerase II transcription elongation past template-encoded arresting sites. Subunit A is transcriptionally active and its transcription activity is strongly enhanced by binding to the dimeric complex of the SIII regulatory subunits B and C (elongin BC complex) (PubMed:7638163). In embryonic stem cells, the elongin BC complex is recruited by EPOP to Polycomb group (PcG) target genes in order generate genomic region that display both active and repressive chromatin properties, an important feature of pluripotent stem cells (By similarity). |
