Description | TDP43, also known as TAR DNA-binding protein 43, plays a crucial role in regulating transcription and splicing of exons (1,2). In healthy conditions, TDP43 is predominantly found in the nucleus. However, in patients with frontotemporal lobar degeneration (FTLD) and amyotrophic lateral sclerosis (ALS), there is an abnormal accumulation of TDP43 in insoluble aggregates. These aggregates are formed due to abnormal ubiquitination, phosphorylation, and cleavage of TDP43, resulting in the generation of carboxy-terminal fragments. These fragments get sequestered as insoluble aggregates in various parts of neurons, including nuclei, perikarya, and neurites (3,4). |

| Application | The recommended dilutions for using this antibody in various assay types are as follows: for Western blotting, dilute the antibody at a range of 1:500 to 1:2000, for immunohistochemistry dilutions at 1:50 to 1:200 are recommended, and when using the antibody in ELISA experiments that involve peptides, dilute the antibody at a range of 1:20000 to 1:40000. It is important to follow the suggested dilutions in order to achieve optimal results when using this antibody in these assay types. |
| Immunogen | A synthesized peptide derived from human TARDBP. |

Concentration | 1mg/ml |
| purification | Peptide affinity chromatography was used to purify the antiserum with the aid of SulfoLink™ Coupling Resin. The resin was utilized to couple peptides to a solid support, enabling the selective binding of the desired antiserum. The resulting purified antiserum was highly specific and effective in its intended application. |

Target | |
Background | DNA and RNA-binding protein which regulates transcription and splicing. Involved in the regulation of CFTR splicing. It promotes CFTR exon 9 skipping by binding to the UG repeated motifs in the polymorphic region near the 3'-splice site of this exon. The resulting aberrant splicing is associated with pathological features typical of cystic fibrosis. May also be involved in microRNA biogenesis, apoptosis and cell division. Can repress HIV-1 transcription by binding to the HIV-1 long terminal repeat. Stabilizes the low molecular weight neurofilament (NFL) mRNA through a direct interaction with the 3' UTR. |
| Tissue specificity | Ubiquitously expressed. In particular, expression is high in pancreas, placenta, lung, genital tract and spleen. |
| Posttranslational modification | Hyperphosphorylated in hippocampus, neocortex, and spinal cord from individuals affected with ALS and FTLDU.Ubiquitinated in hippocampus, neocortex, and spinal cord from individuals affected with ALS and FTLDU.Cleaved to generate C-terminal fragments in hippocampus, neocortex, and spinal cord from individuals affected with ALS and FTLDU. |
