Description

TDP43, also known as TAR DNA-binding protein 43, plays a crucial role in regulating transcription and splicing of exons (1,2). In healthy conditions, TDP43 is predominantly found in the nucleus. However, in patients with frontotemporal lobar degeneration (FTLD) and amyotrophic lateral sclerosis (ALS), there is an abnormal accumulation of TDP43 in insoluble aggregates. These aggregates are formed due to abnormal ubiquitination, phosphorylation, and cleavage of TDP43, resulting in the generation of carboxy-terminal fragments. These fragments get sequestered as insoluble aggregates in various parts of neurons, including nuclei, perikarya, and neurites (3,4).
Furthermore, apart from its role in neurodegenerative disorders, TDP43 also has other important functions. It can inhibit the expression of the HIV-1 gene and regulate the splicing of the CFTR gene (1,5). This highlights the diverse and complex roles played by TDP43 in cellular processes and its significance beyond neurodegenerative diseases.
