Description

Isoform 1 of this protein plays a crucial role in apoptotic morphological changes, including membrane blebbing, cell contraction, and apoptotic bodies formation. Its activation of MAPK8/JNK and nuclear localization of the C-terminally truncated isoform 1 are necessary for this function, which may be associated with the mitochondrial CASP9-dependent death pathway. The JNK MAP kinase pathway is specifically activated by isoform 1 via MKK3 and MKK6 kinases. In addition, it binds to microtubules and affects their organization and stability, independently of its kinase activity. Furthermore, it can prevent MAP3K7-mediated activation of IKKA, and therefore, NF-kappa-B activation, but not that of JNK. Isoform 2, on the other hand, is required for PCDH8 endocytosis, which is involved in homophilic interactions between PCDH8 extracellular domains. This leads to phosphorylation and activation of MAPK14/p38 MAPK, which phosphorylates isoform 2, resulting in PCDH8 endocytosis and CDH2 cointernalization.
