Overview

Description

TAF6L is an essential component of the PCAF complex, a crucial factor involved in histone acetylation within the nucleosome. The PCAF complex, akin to the yeast SAGA complex, comprises TAF5, TAF5L, TAF6, TAF6L, TAF9, TAF10, and TAF12, along with PCAF and PCAF-associated factors (PAFs), which include TADA2L/ADA2, TADA3L/ADA3, and SPT3. As a member of the TAF6 family, TAF6L is an integral part of the STAGA transcription coactivator-HAT complex, which also features SUPT3H, GCN5L2, TAF5L, TAF6L, SUPT7L, TADA3L, TAD1L, TAF10, TAF12, TRRAP, and TAF9. Overall, TAF6L has a key role in facilitating histone acetylation and gene activation as part of the PCAF and STAGA complexes.

Other names

PAF65 alpha, also known as CBP associated factor associated factor 65 kDa subunit 6L or PAF65A, is a protein that is associated with the CBP-associated factor complex. It is also referred to as PCAF associated factor 65 alpha or TAF6-like RNA polymerase II p300/CBP associated factor associated factor 65 kDa subunit 6L. PAF65 alpha plays a critical role in regulating transcription and gene expression. It is involved in diverse cellular processes, such as chromatin remodeling, DNA repair, and cell cycle control. Its interaction with other proteins, such as p300/CBP and PCAF, suggests its involvement in the activation of transcription factors and histone acetyltransferase activity. The exact mechanism of how PAF65 alpha functions in different cellular contexts is still under investigation. However, its association with various transcriptional coactivators and chromatin-modifying enzymes highlights its crucial role in gene regulation.

Application

To ensure accurate results for a variety of assays, recommended dilutions are as follows: western blotting at 1:500-1:2000, immunofluorescence or immunocytochemistry at 1:100-1:500, and ELISA using peptide substrates at 1:20000-1:40000 dilutions. It's important to note that dilution factors may vary based on specific experimental conditions and should be optimized for best results.

Immunogen

A synthesized peptide derived from human TAF6L.

Purification

SulfoLink™ Coupling Resin was employed to perform peptide affinity chromatography and purify the antiserum. The purification process involved the use of the resin to selectively bind and isolate the desired peptides from the mixture. By using this technique, the antiserum was successfully purified to obtain a high-quality product.

Stability & Storage

One should store the product at a temperature of -20 °C for a duration of one year to ensure its longevity. It is important to avoid exposing it to repeated cycles of freezing and thawing to maintain its integrity.

Storage buffer

This Rabbit IgG solution is formulated with phosphate buffered saline at pH 7.4, containing 150mM NaCl and 0.02% sodium azide. Also, it includes 50% glycerol as a stabilizer. It is recommended to store this solution at a temperature of -20°C for maximum stability, and the solution stays stable without degradation for up to 12 months from the date of receipt.

Target

Background

The PCAF complex, which functions as a component of the human version of the yeast SAGA complex, plays a crucial role in efficiently acetylating histones within a nucleosomal context. This complex is highly adept at modifying histones and can be likened to the SAGA complex of yeast.
