Description | This particular gene encodes a member of the sirtuin family of proteins, which are similar to the yeast Sir2 protein. The sirtuin family is made up of four different classes, all of which share a sirtuin core domain. Although the functions of human sirtuins are not yet fully understood, researchers have discovered that yeast sirtuin proteins play a role in regulating epigenetic gene silencing and suppressing rDNA recombination. Some studies suggest that human sirtuins act as intracellular regulatory proteins that possess the ability to perform mono-ADP-ribosyltransferase activity. This particular sirtuin protein falls into class IV of the sirtuin family. This information was provided by RefSeq in July of 2008. |
| Other names | Sirtuin 7, also known as NAD-dependent protein deacetylase sirtuin-7 or regulatory protein SIR2 homolog 7, is a type of NAD-dependent deacetylase. Its function is to remove acetyl groups from proteins and play a role in regulating gene expression and cellular metabolism. Sirtuin 7 is similar to the Silent mating type information regulation 2 homolog (SIR2) found in yeast, and is also referred to as SIR2-like protein 7 (SIR2L7). Its presence has been observed in many organisms, including humans. |

| Application | The recommended dilutions for WB are 1:500-1:2000, for IHC they are 1:50-1:200, and for ELISA (peptide) the dilutions are 1:20000 to 1:40000. It's important to follow these dilutions in order to achieve the most accurate and reliable results in your experiments. Make sure to carefully follow the instructions for your specific assay and sample type, as the optimal dilution may vary depending on your experimental conditions. By using the appropriate dilution, you can ensure that your assays are sensitive and specific, providing you with reliable and meaningful data. |

Background | NAD-dependent protein deacetylase that specifically mediates deacetylation of histone H3 at 'Lys-18' (H3K18Ac). In contrast to other histone deacetylases, displays selectivity for a single histone mark, H3K18Ac, directly linked to control of gene expression. H3K18Ac is mainly present around the transcription start site of genes and has been linked to activation of nuclear hormone receptors. SIRT7 thereby acts as a transcription repressor. Moreover, H3K18 hypoacetylation has been reported as a marker of malignancy in various cancers and seems to maintain the transformed phenotype of cancer cells. These data suggest that SIRT7 may play a key role in oncogenic transformation by suppresses expression of tumor suppressor genes by locus-specific deacetylation of H3K18Ac at promoter regions. Also required to restore the transcription of ribosomal RNA (rRNA) at the exit from mitosis: promotes the association of RNA polymerase I with the rDNA promoter region and coding region. Stimulates transcription activity of the RNA polymerase I complex. May also deacetylate p53/TP53 and promotes cell survival, however such data need additional confirmation. |
