Description

The 26S proteasome, a well-organized multicatalytic proteinase complex, consists of two main components: a 20S core and a 19S regulator. The 20S core comprises four rings, each consisting of seven non-identical subunits. Specifically, two rings consist of alpha subunits, while the other two rings consist of beta subunits. On the other hand, the 19S regulator comprises a base and a lid. The base contains six ATPase subunits and two non-ATPase subunits, while the lid consists of up to ten non-ATPase subunits. These proteasomes are found in abundance throughout eukaryotic cells and are involved in peptide cleavage via an ATP/ubiquitin-dependent mechanism, operating outside the lysosomal pathway.
One important variant of the proteasome, known as the immunoproteasome, plays a critical role in processing class I MHC peptides. In contrast to the standard proteasome, the immunoproteasome replaces the 19S regulator with an alternative regulator called the 11S regulator or PA28. The 11S regulator consists of three subunits: alpha, beta, and gamma. Among these, the beta subunit encoded by this gene is induced by gamma-interferon. When combined with three alpha subunits, the beta subunit forms a heterohexameric ring, contributing to the function of the immunoproteasome.
It is worth noting that several pseudogenes resembling this gene have been identified on chromosomes 4, 5, 8, 10, and 13. These non-functional gene copies do not produce proteins but can still provide insight into the evolutionary history of the genome.
