Description | This particular gene belongs to the specialized DNA polymerase family known as Y. It functions by replicating undamaged DNA but with a lower fidelity compared to other DNA-directed polymerases. Interestingly, it demonstrates a high level of accuracy when it comes to replicating UV-damaged DNA. In the presence of thymine dimers, this polymerase inserts the complementary nucleotides in the newly synthesized DNA, effectively bypassing the DNA lesion and preventing the mutagenic effects caused by UV-induced DNA damage. Additionally, this polymerase has been linked to hypermutation during immunoglobulin class switch recombination. When mutations occur in this gene, it leads to a variant type of xeroderma pigmentosum called XPV. These mutations result in the impaired functionality of the polymerase, compromising its ability to accurately replicate damaged DNA. This information has been sourced from RefSeq, and the content has been arranged in a manner that keeps the original text information intact while ensuring its similarity with the original passage. |
| Other names | FLJ16395 is a gene that encodes DNA polymerase eta, also known as POLH or POLH_HUMAN. Another name for this gene is FLJ21978. It is a crucial enzyme involved in DNA repair and replication processes. POLH is also referred to as RAD30 or RAD30 homolog A, as it shares similarities with the RAD30 gene found in yeast. This gene is especially important in individuals with xeroderma pigmentosum variant type (XPV) disease. XPV is a genetic disorder that impairs the ability of cells to repair UV-induced DNA damage. DNA polymerase eta plays a key role in overcoming this defect. By rearranging the information mentioned above, we can still convey the same details while presenting them in a new structure. |

| Specificity | The POLH Antibody is capable of detecting total POLH levels that are endogenously present in the sample. This indicates that the antibody has the ability to bind with the target molecule in its natural state. To summarize, the POLH Antibody is specifically designed to recognize and measure total POLH protein in a given sample. |

| Application | For Western Blot analysis, the recommended dilution range is 1:500-1:2000. When it comes to Immunohistochemistry, we suggest a dilution range of 1:50-1:200. For ELISA analysis using a peptide, the recommended dilution range is 1:20000-1:40000. It is important to follow the recommended dilution ranges for each application to ensure accurate and reliable results. By following these guidelines, you can achieve optimal performance for your experiments. |

Background | DNA polymerase specifically involved in the DNA repair by translesion synthesis (TLS) (PubMed:10385124, PubMed:11743006, PubMed:24449906). Due to low processivity on both damaged and normal DNA, cooperates with the heterotetrameric (REV3L, REV7, POLD2 and POLD3) POLZ complex for complete bypass of DNA lesions. Inserts one or 2 nucleotide(s) opposite the lesion, the primer is further extended by the tetrameric POLZ complex. In the case of 1,2-intrastrand d(GpG)-cisplatin cross-link, inserts dCTP opposite the 3' guanine (PubMed:24449906). Particularly important for the repair of UV-induced pyrimidine dimers (PubMed:10385124, PubMed:11743006). Although inserts the correct base, may cause base transitions and transversions depending upon the context. May play a role in hypermutation at immunoglobulin genes (PubMed:11376341, PubMed:14734526). Forms a Schiff base with 5'-deoxyribose phosphate at abasic sites, but does not have any lyase activity, preventing the release of the 5'-deoxyribose phosphate (5'-dRP) residue. This covalent trapping of the enzyme by the 5'-dRP residue inhibits its DNA synthetic activity during base excision repair, thereby avoiding high incidence of mutagenesis (PubMed:14630940). Targets POLI to replication foci (PubMed:12606586). |
| Posttranslational modification | Monoubiquitinated by RCHY1/PIRH2; ubiquitination inhibits the ability of PolH to interact with PCNA and to bypass UV-induced lesions. |
| Celluar localization | Cytosol;Nucleus; |
