Description | RBX-box protein 1 (also known as ROC1) plays a vital role as a component of two E3 ubiquitin ligase complexes, namely, the SCF complex and the CBC (VHL) complex. The SCF E3 ligase is activated when RBX1 mediates the neddylation of CUL1 by facilitating the transfer of ubiquitin from E2 to substrates. For ubiquitin ligation, the RING finger domain of RBX1 is crucial. RBX1 belongs to the RBX family, which is conserved in mammals and includes another member, RBX2. RBX1 binds constitutively to CUL2/VHL, while stress-inducible RBX2 binds to CUL5/SOCS. Together, these findings reveal the essential role of RBX1 in the regulation of protein degradation pathways. |
| Other names | RBX1, also known as Ring Box 1, is an E3 ubiquitin-protein ligase that plays an important role in the regulation of protein degradation in cells. This protein is also known as RNF75 and ROC1, and it is involved in the functioning of cullin-RING ligase complexes. The RING box protein 1 is encoded by the RBX1 gene and helps in the binding of ubiquitin to specific targets for their degradation. RBX1 has several aliases like FLJ60363, MGC13357, and MGC1481, indicating its significance in different cell activities. The protein ZYP is one such role for RBX1, and it is known to mediate polyubiquitination of proteins for degradation. Being a RING finger protein, RBX1 has the ability to recruit ubiquitin-conjugating enzymes to its substrate. In simple words, it helps to mark the target proteins for degradation by the proteasome. A better understanding of RBX1 can help in the development of new strategies to treat cancer, as the ubiquitin-proteasome system plays an essential role in cancer development. |

Background | E3 ubiquitin ligase component of multiple cullin-RING-based E3 ubiquitin-protein ligase (CRLs) complexes which mediate the ubiquitination and subsequent proteasomal degradation of target proteins, including proteins involved in cell cycle progression, signal transduction, transcription and transcription-coupled nucleotide excision repair. CRLs complexes and ARIH1 collaborate in tandem to mediate ubiquitination of target proteins, ARIH1 mediating addition of the first ubiquitin on CRLs targets (PubMed:27565346). The functional specificity of the E3 ubiquitin-protein ligase complexes depends on the variable substrate recognition components. As a component of the CSA complex promotes the ubiquitination of ERCC6 resulting in proteasomal degradation. Through the RING-type zinc finger, seems to recruit the E2 ubiquitination enzyme, like CDC34, to the complex and brings it into close proximity to the substrate. Probably also stimulates CDC34 autoubiquitination. May be required for histone H3 and histone H4 ubiquitination in response to ultraviolet and for subsequent DNA repair. Promotes the neddylation of CUL1, CUL2, CUL4 and CUL4 via its interaction with UBE2M. Involved in the ubiquitination of KEAP1, ENC1 and KLHL41. In concert with ATF2 and CUL3, promotes degradation of KAT5 thereby attenuating its ability to acetylate and activate ATM. |
