Description | Trk receptor tyrosine kinases, including TrkA, TrkB, and TrkC, form a family with conserved sequences but are activated by different neurotrophins. TrkA responds to NGF, while TrkB is activated by BDNF or NT4, and TrkC by NT3. These receptors play a crucial role in regulating various physiological processes, such as cell survival, proliferation, neural development, and the growth and patterning of axons and dendrites. In the adult nervous system, Trk receptors also have a role in modulating synaptic strength and plasticity. TrkA specifically affects proliferation and contributes to the development and maturation of the nervous system. The phosphorylation of Tyr490 is essential for the association of Shc and the activation of the Ras-MAP kinase cascade. Moreover, Tyr674/675 residues within the catalytic domain of TrkA reflect its kinase activity through phosphorylation. Certain genetic alterations can lead to ligand-independent receptor dimerization and activation, including point mutations, deletions, and chromosomal rearrangements (chimeras). Notably, TrkA activation has been observed in several malignancies, such as breast, ovarian, prostate, and thyroid carcinomas. In neuroblastomas, the expression of TrkA may serve as a valuable prognostic marker, as it promotes growth arrest and differentiation in cells derived from the neural crest. |
| Other names | BDNF tropomyosine receptor kinase B, also known as TrkB, is a neurotrophic receptor tyrosine kinase type 2 that plays a crucial role in brain development and function. It is encoded by the NTRK2 gene and is involved in various cellular processes, including neuronal survival, differentiation, and synaptic plasticity. TrkB is a receptor for the brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3) growth factors. It belongs to the family of tropomyosin-related kinase receptors and is responsible for transmitting the signaling cascades activated by BDNF and NT-3. This receptor contains an extracellular domain, a transmembrane domain, and an intracellular kinase domain. Mutations or dysregulation in the NTRK2 gene have been associated with obesity, hyperphagia (excessive eating), and developmental delays. This highlights the importance of TrkB signaling in maintaining proper neuronal function and metabolic homeostasis. Overall, TrkB is a critical player in neurotrophin signaling pathways and is involved in various physiological and pathological processes. Understanding its functions and dysregulation may provide insights into the development of therapeutic strategies for neurodevelopmental disorders and metabolic diseases. |

| Specificity | The NTRK2 Antibody is capable of identifying the total amount of NTRK2 protein that is present naturally within an organism. This means that researchers can use this antibody to monitor the levels of this important protein in various tissues and cell types. Furthermore, the antibody specifically detects endogenous levels of NTRK2, which means that it only reacts with the protein when it is expressed by cells naturally rather than as a result of an experimental manipulation. With the help of this valuable tool, scientists can gain a better understanding of the role that NTRK2 plays in various physiological and pathological processes. |

Background | Receptor tyrosine kinase involved in the development and the maturation of the central and the peripheral nervous systems through regulation of neuron survival, proliferation, migration, differentiation, and synapse formation and plasticity. Receptor for BDNF/brain-derived neurotrophic factor and NTF4/neurotrophin-4. Alternatively can also bind NTF3/neurotrophin-3 which is less efficient in activating the receptor but regulates neuron survival through NTRK2. Upon ligand-binding, undergoes homodimerization, autophosphorylation and activation. Recruits, phosphorylates and/or activates several downstream effectors including SHC1, FRS2, SH2B1, SH2B2 and PLCG1 that regulate distinct overlapping signaling cascades. Through SHC1, FRS2, SH2B1, SH2B2 activates the GRB2-Ras-MAPK cascade that regulates for instance neuronal differentiation including neurite outgrowth. Through the same effectors controls the Ras-PI3 kinase-AKT1 signaling cascade that mainly regulates growth and survival. Through PLCG1 and the downstream protein kinase C-regulated pathways controls synaptic plasticity. Thereby, plays a role in learning and memory by regulating both short term synaptic function and long-term potentiation. PLCG1 also leads to NF-Kappa-B activation and the transcription of genes involved in cell survival. Hence, it is able to suppress anoikis, the apoptosis resulting from loss of cell-matrix interactions. May also play a role in neutrophin-dependent calcium signaling in glial cells and mediate communication between neurons and glia. |
| Tissue specificity | Isoform TrkB is expressed in the central and peripheral nervous system. In the central nervous system (CNS), expression is observed in the cerebral cortex, hippocampus, thalamus, choroid plexus, granular layer of the cerebellum, brain stem, and spinal cord. In the peripheral nervous system, it is expressed in many cranial ganglia, the ophthalmic nerve, the vestibular system, multiple facial structures, the submaxillary glands, and dorsal root ganglia. Isoform TrkB-T1 is mainly expressed in the brain but also detected in other tissues including pancreas, kidney and heart. Isoform TrkB-T-Shc is predominantly expressed in the brain. |
| Posttranslational modification | Phosphorylated. Undergoes ligand-mediated autophosphorylation that is required for interaction with SHC1 and PLCG1 and other downstream effectors. Isoform TrkB-T-Shc is not phosphorylated.Ubiquitinated. Undergoes polyubiquitination upon activation; regulated by NGFR. Ubiquitination regulates the internalization of the receptor (By similarity). |
