Description

The Trk receptor tyrosine kinases family comprises TrkA, TrkB, and TrkC. Although sharing a highly conserved sequence, these receptors are activated by distinct neurotrophins: TrkA by NGF, TrkB by BDNF or NT4, and TrkC by NT3, respectively. Neurotrophin signaling via these receptors plays a crucial role in mediating several physiological processes such as cell survival, neural development, proliferation, dendrite growth, and axon patterning. In the adult nervous system, the Trk receptors are involved in the regulation of synaptic strength and plasticity. TrkA, in particular, is essential for the development and maturation of the nervous system as well as for regulating cell proliferation. It has been found that phosphorylation at Tyr490 is crucial for Shc association, which leads to the activation of the Ras-MAP kinase cascade. Additionally, Tyrosine residues at positions 674/675 within the catalytic domain reflect TrkA kinase activity. Aberrant TrkA activation caused by point mutations, deletions, and chromosomal rearrangements, may lead to ligand-independent receptor dimerization. Moreover, TrkA is frequently activated in different malignancies including ovarian, breast, prostate, and thyroid carcinomas. Interestingly, TrkA expression appears to be a favorable prognostic marker for neuroblastomas. This is because TrkA signaling leads to growth arrest and differentiation of cells derived from the neural crest, which neuroblastomas originate from.

Can also bind and be activated by NTF3/neurotrophin-3. However, NTF3 only supports axonal extension through NTRK1 but has no effect on neuron survival (By similarity). Upon dimeric NGF ligand-binding, undergoes homodimerization, autophosphorylation and activation (PubMed:1281417). Recruits, phosphorylates and/or activates several downstream effectors including SHC1, FRS2, SH2B1, SH2B2 and PLCG1 that regulate distinct overlapping signaling cascades driving cell survival and differentiation. Through SHC1 and FRS2 activates a GRB2-Ras-MAPK cascade that regulates cell differentiation and survival. Through PLCG1 controls NF-Kappa-B activation and the transcription of genes involved in cell survival. Through SHC1 and SH2B1 controls a Ras-PI3 kinase-AKT1 signaling cascade that is also regulating survival.
