Overview | |
Description | MAST2 is a protein that provides a link between the dystrophin/utrophin network and microtubule filaments through the syntrophins. The affinities of DMD or UTRN for associated proteins may be altered by phosphorylation. This protein has a role in spermatid maturation as part of a multi-protein complex. Additionally, MAST2 regulates the synthesis of IL-12 in macrophages in response to lipopolysaccharides by forming a complex with TRAF6, which inhibits TRAF6 NF-kappaB activation. Two different isoforms of this protein are produced in human beings via alternative splicing. |
Other names | Microtubule-associated serine/threonine-protein kinase 2, also known as MAST2 or MAST205, is an enzyme encoded by several gene names, including KIAA0807, FLJ39200, RP4-533D7.1 and OTTHUMP00000009697. This kinase plays a crucial role in microtubule regulation by phosphorylating various microtubule-associated proteins, which affects cell morphology, motility and division. MAST2 is mainly expressed in the nervous system and testis and has been implicated in neuronal development and spermatogenesis. Due to its essential functions, MAST2 is a potential therapeutic target for diseases involving abnormal microtubule dynamics, such as cancer and neurodegenerative disorders. |

Application | To properly utilize the WB 1:500-1:2000, IF/ICC 1:100-1:500, and ELISA(peptide) 1:20000-1:40000 techniques, it is important to understand their specific dilution ranges. For western blotting, a dilution range of 1:500-1:2000 is recommended, while IF/ICC should be diluted between 1:100-1:500. For ELISA using peptides, the appropriate dilution range is 1:20000-1:40000. These dilutions are crucial in ensuring reliable and accurate results in your experiments. By following the correct dilution ranges, you can optimize your experiments and obtain meaningful data. |

Stability & Storage | To ensure optimum storage conditions, keep the product at a temperature of -20°C for up to a year. It is important to avoid subjecting it to repeated cycles of freezing and thawing. Therefore, be mindful of storage practices when handling this product. |
Storage buffer | The stable storage conditions for Rabbit IgG are as follows: it should be stored at -20 °C in phosphate buffered saline with a pH of 7.4. The solution should contain 150mM NaCl, 0.02% sodium azide, and 50% glycerol. The Rabbit IgG is expected to remain stable for a period of 12 months starting from the date of receipt. |
Target | |
Background | The syntrophins appear to establish a connection between the dystrophin/utrophin network and microtubule filaments. The affinities of DMD or UTRN for associated proteins may be influenced by their phosphorylation. During spermatid maturation, it plays a role within a complex comprising multiple proteins. By interacting with TRAF6, it forms a complex that hampers NF-kappa-B activation, ultimately inhibiting lipopolysaccharide-induced IL-12 synthesis in macrophages. |
Tissue specificity | Abundant in the testis. |
Posttranslational modification | Phosphorylated and ubiquitinated. N-terminal ubiquitination leads to degradation of MAST2 by proteasome-mediated proteolysis. N-terminal phosphorylation appears to be a prerequisite for ubiquitination (By similarity). |
Celluar localization | Cytoskeleton;Plasma Membrane; |
UniPort | Q6P0Q8 |
