Description

LT-α, also called TNF-β, is a protein belonging to the TNF superfamily (1). It is expressed by various immune cells like NK cells, T cells, B cells, and lymphoid tissue-inducer cells (1). LT-α can exist as a soluble homotrimer or form membrane bound heterotrimers with lymphotoxin-β (LT α1β2 or LT α2β1), which can be cleaved from the cell surface by matrix metalloproteases (1,2). The soluble form of LT-α binds to TNFR1/TNFR2 and activates the canonical NF-κB pathway (1). On the other hand, LT α1β2 heterodimers bind to the LTβR receptor and activate the noncanonical NF-κB pathway (1). Consequently, LT-α and TNF-α exhibit similar functions. Soluble LT-α and LT α1β2 are crucial in lymphangiogenesis (3). The LT α1β2/LTβR axis plays a vital role in the development of lymphoid tissue (1,3).

Other namesAlpha lymphotoxin (LTalpha) is a member of the tumor necrosis factor (TNF) superfamily, specifically known as TNF superfamily member 1. It is also referred to as LT, LT-alpha, Lta, or lymphotoxin-alpha. TNF superfamily member 1, also known as TNF B or TNF-beta, is another name for LTalpha. TNF, lymphocyte-derived, TNFB, TNFB_HUMAN, TNFbeta, TNFSF1, TNLG1E, and tumor necrosis factor ligand superfamily member 1E are all aliases for LTalpha. This protein, known as tumor necrosis factor ligand 1E, plays a significant role in immune responses and inflammation. Thus, various terms exist to describe LTalpha due to its importance in these biological processes.

ApplicationWestern blot (WB) is typically performed using dilutions ranging from 1:500 to 1:2000. Similarly, for immunohistochemistry (IHC), the recommended dilutions are between 1:50 and 1:200. On the other hand, ELISA assays using peptide antigens usually require higher dilutions, ranging from 1:20000 to 1:40000. These dilution ranges ensure optimal signal detection and accurate quantification of target proteins or antigens in the respective assays.

Background

Cytokine that in its homotrimeric form binds to TNFRSF1A/TNFR1, TNFRSF1B/TNFBR and TNFRSF14/HVEM. In its heterotrimeric form with LTB binds to TNFRSF3/LTBR. Lymphotoxin is produced by lymphocytes and cytotoxic for a wide range of tumor cells in vitro and in vivo.
