Description | An important player in the establishment of tight junctions among epithelial cells, this protein plays a key role in early forms of cell junctions and recruits a protein called PARD3. The interaction between PARD6-PARD3 complex may prevent PARD3 from bonding with JAM1, which subsequently prevents the formation of tight junctions. It also aids in regulating the mobility of monocytes across the epithelial barrier. Additionally, it has been shown to activate platelets. In the case of infection with orthoreovirus, this protein serves as a receptor for the virus. |
| Other names | CD321, also known as junctional adhesion molecule 1 (JAM1), is a protein that plays a crucial role in cell adhesion and migration. It is also known as F11 receptor and platelet adhesion molecule 1 (PAM1). The CD321 antigen is expressed on a wide range of cells, including endothelial cells, leukocytes, and platelets. JAM1 is involved in the maintenance of endothelial barrier function, leukocyte recruitment, and platelet activation and aggregation. It interacts with other proteins in the junctional complex, such as ZO-1, Afadin, and PAR-3, to regulate cellular processes. JAM1 has been identified as a potential therapeutic target in several diseases, including cancer, autoimmune disorders, and inflammation. Blocking JAM1-mediated interactions has been shown to inhibit tumor growth and metastasis, reduce inflammation, and improve outcomes in animal models of disease. The homolog of JAM1 in mice is also known as junctional adhesion molecule A (JAMA) or JAM-A. It shares high sequence similarity with human JAM1 and plays similar roles in cell adhesion and migration. In conclusion, CD321/JAM1 is a versatile protein that is involved in a range of cellular processes and has potential therapeutic applications in various diseases. |

Background | Seems to play a role in epithelial tight junction formation. Appears early in primordial forms of cell junctions and recruits PARD3 (PubMed:11489913). The association of the PARD6-PARD3 complex may prevent the interaction of PARD3 with JAM1, thereby preventing tight junction assembly (By similarity). Plays a role in regulating monocyte transmigration involved in integrity of epithelial barrier (By similarity). Ligand for integrin alpha-L/beta-2 involved in memory T-cell and neutrophil transmigration (PubMed:11812992). Involved in platelet activation (PubMed:10753840). |
| Tissue specificity | Expressed in endothelium, epithelium and leukocytes (at protein level). |
| Posttranslational modification | N-glycosylated. |
| Celluar localization | Extracellular region or secreted;Plasma Membrane; |
