Background	This protein acts as a mediator between various growth factor receptors that have tyrosine kinase activity, including insulin receptor, IGF1R, and FGFR1. It forms a connection with a complex network of signaling molecules inside cells that contain SH2 domains. Its main role is in the mitogenic signaling pathway of IGF1R. During insulin stimulation, it promotes the AKT1 signaling pathway and facilitates BAD phosphorylation without affecting RPS6KB1 activation or apoptosis inhibition. When interacting with GRB2, it enhances the activity of mitogen-activated protein kinase in response to insulin. It may also play a part in nonreceptor tyrosine kinase signaling in myoblasts. Additionally, it plays a crucial role in the proliferation and differentiation of hepatoblastoma cells by activating EPHB2 upon IGF1 stimulation. In terms of signal transduction, it is involved in the response to insulin, as well as to a lesser extent, IL4 and GH, with respect to mitogenesis.
