Description

IL-6, also known as interleukin-6, is a versatile cytokine that is produced by various cells in response to infection, trauma, and immune challenges (PMID: 8274730). Its signaling activity relies on a receptor complex composed of two different membrane-bound glycoproteins: the IL-6-binding subunit (IL-6R or CD126 or gp80) and the signal-transducing element (gp130 or CD130), weighing 80-kDa and 130-kDa, respectively. Upon binding of IL-6 to the IL-6R on the cell membrane, the IL-6/IL-6R complex associates with gp130, initiating receptor activation (PMID: 9716487).
The distribution of IL-6R and gp130 is characterized by variation. While gp130 is expressed in nearly all organs, IL-6R expression is primarily observed in hepatocytes and specific leukocyte subtypes such as monocytes, neutrophils, T cells, and B cells (PMID: 11149892). Interestingly, the soluble form of IL-6R has also been detected in human serum and urine. These findings highlight the involvement of IL-6 in various physiological processes and its role as a communicator in immune responses and inflammatory conditions.
