Description

HEXIM1 is a crucial transcriptional regulator that inhibits general RNA polymerase II transcription. By collaborating with 7SK snRNA, it effectively sequesters P-TEFb in a large and inactive 7SK snRNP complex, thus restraining RNA polymerase II phosphorylation and subsequent transcriptional elongation. Moreover, this protein has been reported to regulate many transcriptional activities, including NF-kappa-B, ESR1, NR3C1, and CIITA. HEXIM1 forms homooligomer and heterooligomer with HEXIM2, which is probably dimeric. It is also an essential component of the 7SK snRNP complex that consists of P-TEFb, HEXIM1, HEXIM2, BCDIN3, SART3 proteins, and 7SK and U6 snRNAs. Besides, it interacts with the N-CoR complex through NCOR1.

Cardiac lineage protein 1, also known as CLP1 or EDG1, is a hexamethylene bis acetamide-inducible protein that plays a role in the regulation of gene expression. It is encoded by the estrogen down-regulated gene 1, or EDG1, and is also referred to as FLJ13562 or HIS1. CLP1 is a member of the Menage a quatre protein family, and is involved in the formation of a transcriptionally active complex that includes the positive transcription elongation factor b, or P-TEFb. This complex is important for the regulation of RNA polymerase II-mediated transcription and is inducible by hexamethylene bis acetamide, or HMBA. CLP1 is therefore considered to be a key mediator of transcriptional regulation and has potential as a therapeutic target in cancer and other diseases.
