Description | The transfer of glutathione's glutamyl moiety to various amino acids and dipeptide acceptors is facilitated by an enzyme encoded by this gene. Comprising a heavy chain and a light chain, both derived from a single precursor protein, this enzyme is prevalent in tissues involved in absorption and secretion. Belonging to the gamma-glutamyltransferase protein family, which comprises numerous incompletely characterized members, some of these enzymes may even be pseudogenes. Specifically, this gene falls under the type I gamma-glutamyltransferase classification. Interestingly, multiple alternatively spliced variants encoding the same protein have been discovered (RefSeq, Oct 2008). |
| Other names | This is a list of gene names associated with Gamma glutamyltranspeptidase, a key enzyme involved in the metabolism of glutathione. The gene names include CD224, D22S672, D22S732, Gamma glutamyltranspeptidase 1, GGT, GGT1, and GGT1_HUMAN. Other aliases associated with this gene include Glutathione hydrolase 1, Leukotriene C4 hydrolase, MGC96892, MGC96904, MGC96963, OTTHUMP00000028921, and OTTHUMP00000197959. These names represent the different ways in which this gene has been identified across various studies and databases. |

| Specificity | The GGT1 Antibody is capable of detecting total GGT1 in the body at its natural levels. This means that it can identify the substance without altering its properties. To create similar content, the original text can be rearranged to convey the same meaning. |
| Species Reactivity | Human;Mouse;Rat |
| Antigen | GGT1 |
| Application | The dilutions for western blotting range from 1:500 to 1:2000, for immunohistochemistry from 1:50 to 1:200, for immunofluorescence/immunocytochemistry from 1:100 to 1:500, and for ELISA assays using peptides from 1:20000 to 1:40000. These dilutions are essential in various laboratory techniques to ensure optimal detection and quantification of specific proteins or antigens. Properly adjusting the dilutions is crucial for obtaining accurate and reliable results. |

Background | Cleaves the gamma-glutamyl bond of extracellular glutathione (gamma-Glu-Cys-Gly), glutathione conjugates, and other gamma-glutamyl compounds. The metabolism of glutathione releases free glutamate and the dipeptide cysteinyl-glycine, which is hydrolyzed to cysteine and glycine by dipeptidases. In the presence of high concentrations of dipeptides and some amino acids, can also catalyze a transpeptidation reaction, transferring the gamma-glutamyl moiety to an acceptor amino acid to form a new gamma-glutamyl compound. Initiates extracellular glutathione (GSH) breakdown, provides cells with a local cysteine supply and contributes to maintain intracellular GSH level. It is part of the cell antioxidant defense mechanism. Isoform 3 seems to be inactive. |
| Tissue specificity | Detected in fetal and adult kidney and liver, adult pancreas, stomach, intestine, placenta and lung. Isoform 3 is lung-specific. There are several other tissue-specific forms that arise from alternative promoter usage but that produce the same protein. |
| Posttranslational modification | N-glycosylated on both chains. Contains hexoses, hexosamines and sialic acid residues. Glycosylation profiles tested in kidney and liver tissues reveal the presence of tissue-specific and site-specific glycan composition, despite the overlap in composition among the N-glycans. A total of 36 glycan compositions, with 40 unique structures are observed. Up to 15 different glycans are observed at a single site, with site-specific variation in glycan composition. The difference in glycosylation profiles in the 2 tissues do not affect the enzyme activity.Cleaved by autocatalysis into a large and a small subunit and the autocatalytic cleavage is essential to the functional activation of the enzyme. |
| Celluar localization | Extracellular region or secreted;Plasma Membrane; |
