Description | Type II collagen, known as COL2A1, is a protein that plays a crucial role in the development of cartilaginous tissues. It's indispensable in the embryonic formation of the skeleton and enabling linear growth. This protein is also critical in the cartilage's ability to withstand compressive forces. COL2A1 belongs to the fibrillar collagen family and is made up of homotrimers of alpha 1(II) chains. Three isoforms of this protein are produced through alternative splicing in humans. |
| Other names | Type II collagen, also known as alpha 1 type II collagen, plays a crucial role in maintaining healthy cartilage. It is encoded by the COL2A1 gene and is essential for the structural integrity of various connective tissues. In addition to its role in cartilage, type II collagen is also involved in the formation and maintenance of other tissues like the inner ear and the vitreous humor of the eye. Mutations in the COL2A1 gene can result in various genetic disorders, including Stickler syndrome, spondyloepiphyseal dysplasia congenita (SEDC), and achondrogenesis type II. These conditions are characterized by abnormal skeletal development and joint abnormalities. Research has shown that type II collagen may have therapeutic potential in the treatment of certain musculoskeletal disorders, such as osteoarthritis. The use of type II collagen supplements has been suggested to support cartilage health and reduce joint pain and inflammation. Understanding the structure and function of type II collagen is crucial for advancing our knowledge of connective tissue disorders and developing new treatment strategies. Ongoing research in this field aims to unravel the complex mechanisms underlying collagen synthesis, assembly, and regulation, with the ultimate goal of improving the diagnosis, prevention, and management of collagen-related diseases. |

| Application | For western blotting, the recommended dilution range is between 1:500 to 1:3000. When it comes to immunohistochemistry, the suggested dilution is between 1:50 to 1:200. For IF/ICC, the recommended dilution range is between 1:100 to 1:500. As for ELISA (peptide), the suggested dilution is between 1:20000 to 1:40000. To summarize, these are the appropriate dilution ranges for each application. |

Background | Type II collagen is specific for cartilaginous tissues. It is essential for the normal embryonic development of the skeleton, for linear growth and for the ability of cartilage to resist compressive forces. |
| Tissue specificity | Isoform 2 is highly expressed in juvenile chondrocyte and low in fetal chondrocyte. |
| Posttranslational modification | Probably 3-hydroxylated on prolines by LEPREL1 (By similarity). Proline residues at the third position of the tripeptide repeating unit (G-X-P) are hydroxylated in some or all of the chains. Proline residues at the second position of the tripeptide repeating unit (G-P-X) are hydroxylated in some of the chains. The N-telopeptide is covalently linked to the helical COL2 region of alpha 1(IX), alpha 2(IX) and alpha 3(IX) chain. The C-telopeptide is covalently linked to an another site in the helical region of alpha 3(IX) COL2. |
