PTPN13 is a member of the protein-tyrosine phosphatase family and belongs to the non-receptor class subfamily. It plays a crucial role in negatively regulating FAS-induced apoptosis and the pro-apoptotic signaling mediated by NGFR. PTPN13 interacts with TRIP6 and TNFRSF6 (Fas receptor) through its second PDZ domain, while its third PDZ domain interacts with the C-terminal SVP motif of NGFR. This glycoprotein serine protease, located on the cell surface, is involved in various cellular processes such as tissue remodeling, fibrosis, wound healing, inflammation, and tumor growth. It plays a crucial role in extracellular matrix degradation. Both the soluble and plasma membrane forms of this protease exhibit endopeptidase activity, specifically targeting amino acid sequences with an Ala/Ser-Gly-Pro-Ser/Asn/Ala consensus. It can degrade proteins like alpha-2-antiplasmin and SPRY2, as well as gelatin and heat-denatured type I collagen. However, it does not act on native collagen type I and IV, vibronectin, tenascin, laminin, fibronectin, fibrin, or casein. Interestingly, it also possesses dipeptidyl peptidase activity, breaking down dipeptides with a proline residue at the penultimate position, with a preference for certain amino acid combinations. Neuropeptide hormones like neuropeptide Y (NPY), peptide YY (PYY), substance P (TAC1), and brain natriuretic peptide 32 (NPPB) serve as natural substrates for its dipeptidyl peptidase activity. In association with DPP4, PLAUR, or integrins, the plasma membrane form of this protease contributes to pericellular proteolysis of the extracellular matrix, thereby promoting cell adhesion, migration, and invasion. It is also involved in tissue remodeling during development and wound healing. In malignant melanomas, it enhances cell invasiveness through the extracellular matrix. Furthermore, it promotes tumor growth progression by increasing angiogenesis, collagen fiber degradation, apoptosis, and inhibiting the immune system's antitumor response. However, it acts as a tumor suppressor in melanocytic cells, regulating cell proliferation and survival independently of its serine protease activity.
