Description

This protein is a histone methyltransferase that specifically targets the lysine 9 residue of histone H3 (H3K9). It is responsible for adding one or two methyl groups (mono- and dimethylation) to this residue in regions of euchromatin. The addition of these methyl groups, known as H3K9me1 and H3K9me2, respectively, serves as an epigenetic mark for transcriptional repression. This mark recruits proteins called HP1, which bind to the methylated histones and contribute to gene silencing.
In addition to its role in H3K9 methylation, this protein also weakly methylates the lysine 27 residue of histone H3 (H3K27me). It is involved in DNA methylation, but interestingly, its histone methyltransferase activity is not required for this function. This suggests that the two activities, DNA methylation and histone methylation, operate independently.
The protein is likely guided to histone H3 by various DNA-binding proteins such as E2F6, MGA, MAX, and/or DP1. During the G0 phase of the cell cycle, it is believed to contribute to the repression of MYC- and E2F-responsive genes, potentially playing a role in the transition from G0 to G1.
Beyond its involvement in histone methylation, this protein also methylates proteins other than histones. One of its targets is the tumor suppressor protein p53/TP53, specifically at the lysine 373 residue, where it adds two methyl groups (dimethylation).

It is responsible for methylating histone H3 at lysine 9, thereby regulating gene expression and chromatin remodeling. EHMT1 is also sometimes referred to as G9a-like protein 1 or GLP1 due to its close similarity to the G9a histone methyltransferase. Mutations in EHMT1 and its dysregulation have been linked to various human diseases and disorders including schizophrenia, intellectual disability, and cancer. This protein is highly important for maintaining proper cellular function and growth, making it a topic of intense research in both basic and clinical sciences.
